中国野桑蚕mtDNA COⅡ-ATPase 6基因区域的序列测定及分析

摘　　要：对中国山东青州野桑蚕mtDNACOⅡ-ATPase6基因区域进行了序列测定与分析。结果表明在所测定的1.9kb左右的片段中,含有COⅠ基因的部分序列、tRNA-Leu基因、COⅡ基因、tRNA-Lys基因、tRNA-Asp基因、ATPase8基因、ATPase6基因和部分COⅢ基因,该片段的基因组织结构与其他家蚕/野桑蚕的基本一致,显示蚕类线粒体基因排列的保守性。COⅡ、ATPase8、ATPase6三个结构基因不同家蚕/野桑蚕之间的比对分析表明,它们在核苷酸水平、氨基酸水平的同源性很高。结构基因密码子与其他蚕类的相似;3个tRNA基因(tRNA-Leu、tRNA-Lys、tRNA-Asp)的碱基错配率高,TψC环缺乏相对保守的T-ψ-C-Pu-A序列,各臂的碱基配对数、各环碱基数目变化较大。以COⅡ、ATPase8、ATPase6基因的核苷酸序列进行家蚕/野桑蚕的分子进化分析表明,家蚕同中国野桑蚕的亲缘关系最近,是家蚕起源于中国野桑蚕的又一证据。Sequence analysis showed the fragment from COⅡ to ATPase 6 on mitochondrial DNA (mtDNA) of Bombyx mandarina.Qingzhou strain contains partial COⅠ, tRNA^(Leu), COⅡ, tRNA^(Lys), tRNA^(Asp), ATPase 8, ATPase 6 and part COⅢ genes, is about 1.9kb. The similarity of the fragments with other silkworm/wild silkworm showed the conserved arrangement of genes in Bombyx mtDNA. Aligment analysis of genes of COⅡ, ATPase 8 and ATPase 6 respectively showed that there are high homology on the level of nucleotide acid and amino acid among different silkworm/wild silkworm. The codons of the structural genes are alike to others of Bombyx mtDNA. The characters of 3 tRNA (tRNA^(Leu), tRNA^(Lys), tRNA^(Asp)) genes are the high mismatch of bases, lacking corresponding T-ψ-C-Pu-A structure of TψC loop, having large variations in the number of matching basepairs of stems and in the number of base of loops. Molecular evolution analysis of Bombyx based on genes of COⅡ, ATPase 8, ATPase indicated Bombyx mori have the closest relatives with Chinese Bombyx mandarina and come up with another clue that Bombyx mori derived from Chinese Bombyx mandarina.
