摘  要:目的:研究视黄酸(retinoic acid,RA)对体外培养的鸡巩膜成纤维细胞的生长及其超微结构的影响。方法:取第2~3代鸡巩膜成纤维细胞于96孔板中培养24h至贴壁后,分为A组(对照组)和各种终浓度梯度的RA实验组:B组(6×10~7mol/L)、C组(6×10~6mol/L)、D组(6×10~5mol/L)和E组(6×10^(-4)mol/L)分别作用24,48,72h,以MTT比色法检测RA对鸡巩膜成纤维细胞增殖的影响。另分别取对照组及6×10~5mol/LRA组对数生长期2代细胞铺片采用透射电镜观察其超微结构的改变。结果:与对照组相比,6×10~7mol/LRA对鸡巩膜成纤维细胞增殖没有明显的抑制作用(P >0.05);6×10~5mol/L及作用24h以上6×10~6mol/LRA明显抑制体外培养的鸡巩膜成纤维细胞的增殖(P<0.01)。6×10~4mol/LRA组细胞普遍变性崩解,抑制作用最强。与对照组比较,RA组细胞生长明显受抑制,胞质水肿,部分线粒体水肿空泡变,内质网扩张,细胞器呈变性改变。结论:RA对体外培养的鸡巩膜成纤维细胞的增殖有明显的抑制作用,并引起其超微结构的改变。可能是近视发生机制中的一个重要成分。AIM: To evaluate the effect of retinoic acid (RA) on chick scleral fibroblasts proliferation and ultrastructural changes in vitro.· METHODS: Early passages (second-third passage) of scleral fibroblasts were placed in 96-well tissue culture plates and incubated for 24h, then were exposed to different concentrations of RA (0, 6×10-7mol/L, 6×10-6mol/L, 6×10-5mol/L and 6×10-4mol/L) for 24, 48 and 72h respectively. Effects of RA on chick scleral fibroblasts proliferation were evaluated by MTT colorimetric assay. The effect of RA on chick scleral fibroblasts ultrastructure was observed by transmission electron microscope.· RESULTS: RA at the concentration of 6×10-7mol/L did not inhibit the proliferation of chick scleral fibroblasts (P > 0.05). Chick scleral fibroblasts proliferation exposed to RA at the concentration of 6×10-5mol/L, 6×10-6mol/L for more than 24h and 6×10-4mol/L were significantly inhibi- ted, showing a dose-dependent manner (P <0.01). There were significant ultrastructural changes in chick scleral fibroblasts under the effect of RA.· CONCLUSION: RA inhibits chick scleral fibroblasts proliferation significantly in vitro and causes significant ultrastructural changes. It may play an important role in the pathogenesis of myopia.·
