摘  要:目的:研究正常人骨髓成纤维样基质细胞HFCL对 急性髓细胞白血病HL 60细胞cxcr4基因表达的影响及意义. 方法:采用流式细胞仪检测细胞周期,NBT还原实验和 CD11b,CD14,CD13,CD33细胞表面抗原的测定作为细胞分 化的指标;采用半定量RT PCR,Northernblot检测cxcr4基因 的表达.结果:HL 60细胞与HFCL细胞共培养后,NBT阳性 细胞增高,CD11b和CD14的表达增多,并有明显的统计学意 义;而且cxcr4mRNA的表达下调.结论:HFCL细胞能诱导 HL 60细胞部分向单核细胞分化,并使趋化因子受体cxcr4表 达下调.AIM: To study the effect of human bone marrow fibroblastoid stromal cells (HFCL) on the expression of cxcr4 gene in acute myeloid leukemia HL-60 cells. METHODS: The cell cycle was analyzed by flow cytometry and the cell differentiation was determined by morphologic observation of the ability of NBT cells and flow cytometric detection of expression of CD11b, CD14, CD13 and CD33. The expression of cxcr4 mRNA was tested by semi-quantitative RT-PCR and Northern blot. RESULTS: The NBT positive cells and the expression of CD11b and CD14 increased. After in direct contact with HFCL cells for 96 h, the mRNA expression levels of cxcr4 genes were down-regulated. CONCLUSION: HFCL cells can induce monocytic differentiation in HL-60 cells and weaken the ability of metastasis and invasion of HL-60 cells.
