摘 要:目的 探讨γ 干扰素与维拉帕米联合应用逆转人肺癌细胞系HTB-56R多药耐药及机制。方法 采用噻唑蓝(MTT)比色法来研究钙通道阻止剂维拉帕米和人类重组γ- 干扰素对顺铂(DDP)细胞毒性在人类肺癌细胞系HTB -56R上的影响。采用透射电镜、DNA琼脂糖凝胶电泳和流式细胞仪来研究HTB -56R细胞的凋亡。结果 γ- 干扰素与维拉帕米能部分恢复 HTB- 56R对 DDP的敏感性。联合作用后观察到HTB- 56R细胞凋亡的证据,如核固缩、琼脂糖凝胶电泳呈梯形条带,并增加细胞凋亡率。联合γ -干扰素与维拉帕米比单独使用一种能使更多的细胞发生凋亡。结论 γ- 干扰素与维拉帕米能逆转HTB- 56R的耐药性,联合使用有更好的效果。Objective To study the reversion of drug resistance by verapamil (VPL) combined with INF-γ. Methods The MTT colorimetric assay was used to study the effect of VPL and INF-γ on the cytotoxicity of DDP in human lung cancer cell line HTB-56R in vitro.The apoptosis of HTB-56R cells was studied with transmission electron microscope(TEM),DNA ladder by gel electrophoresis and flow cytometic analysis.Results The results showed that sensitivity to DDP was partly reestablished by VPL or INF-γ.Combining use of VPL,INF-γ and DDP,the evidence of apoptosis of HTB-56R cells such as apoptotic bodies and DNA ladder,was observed by TEM and gel electrophoresis.VPL or INF-γ could increase apoptotic rate shown by flow cytometic analysis.The apoptosis rate in combination group of VPL and INF-γ was higher than that in either of the two alone.Conclusion VPL and INF-γ could revert the multidrug resistance of HTB-56R.Combining use of VPL and INF-γ was better than either of the two alone.The reversion mechanism of VPL and INF-γ may be relevant to the increase of apoptosis.
