摘  要:【目的】研究丹参酮Ⅱa (TanⅡa)对大鼠缺氧缺血性脑损伤 (HIBD)脑细胞内游离钙 [(Ca2 + )i]的影响。【方法】利用HIBD动物模型 ,将 30只新生SD大鼠随机均分为正常对照组 ,HIBD组和HIBD +TanⅡa组 ,于低氧处理之前予以 1μg/ gTanⅡa腹腔注射 ,低氧结束时取大鼠脑组织并分离左、右 (右侧为HIBD侧 )大脑半球脑组织制成单细胞悬液 ,以Fura 2标记、以荧光扫描仪测定并比较两侧大脑半球 [(Ca2 + )i]比值。【结果】HIBD +TanⅡa组HIBD侧与对侧大脑半球 [(Ca2 + )i]比值较HIBD组降低 2 4 .9% ,两者间差异有显著性 (P <0 .0 5 )。【结论】TanⅡa可降低缺血缺氧损伤后极早期 [(Ca2 + )i]聚集。临床上丹参对HIE的治疗作用可能与TanⅡa该作用有关。Objective To study the effects of Danshentong IIa ( DanⅡa ; a ketone prepared from Radix salviae miltiorrhizae) on the brain intracellular free calcium [(Ca 2+ )i] in hypoxia-ischemic brain damage (HIBD) of neonatal rats.Adopting HIBD animal model, 30 neonatal SD rats were randomly divided into normal control, HIBD and HIBD plus DanⅡa groups. One ug/g DanⅡa was given intraperitoneally before the hypoxia. By the end of hypoxia, animals were sacrificed by decapitation. The brain tissues were collected, then the left and right hemisphere (i.e.the side subjected to HIBD) were separated to prepare single cell suspension. The [(Ca 2+ )i] ratio between bilateral hemispheres was estimated by Fura-2 labeling and fluorescence scanner and further compared .The [(Ca 2+ )i] ratio of HIBD side hemisphere to opposite side hemisphere in HIBD plus DanⅡa group was 24.9% less than that in HIBD group, showing significant difference between both (P<0.05).[Conclusion]DanⅡa can reduce [(Ca 2+ )i] accumulation in the earliest stage after HIBD, which may be related to the neuro-protective effect of Radix salviae miltiorrhizae on hypoxia-ischemic encephalopathy (HIE).
