摘 要:目的 探讨辐射损伤后HR2 4L修复蛋白复合物的形成 ,为HR2 4L参与切除修复和重组修复提供生化证据。方法 利用质粒共转染和免疫共沉淀技术检测HR2 4L复合物中关键修复蛋白的存在及其在辐射损伤后的变化。结果 2 0J m2 紫外线照射损伤后 2h ,可检测到HR2 4L ,复合物中存在有XPA。 2 0Gyγ射线照射后 2h ,检测到Ku70参与HR2 4L复合物的形成 ,照射后 8h可见Rad5 2存在。结论 辐射损伤后 ,切除修复蛋白和重组修复蛋白参与HR2 4L复合物形成的生化证据 ,进一步支持HR2 4L参与多种修复途径的论点。Objective To investigate HR24L repair complex after DNA damage induced by radiation and provide biochemical evidence of the participation of HR24L in DNA excision repair and recombination repair. Methods Cotransfection and coimmunoprecipitations were employed to detect the presence of key repair proteins in HR24L compex. Results XPA was present in HR24L precipitations 2 h after 20 J/m+2 UVirradiation.When cells were irradiated with 20 Gy γrays,Ku70 was precipitated with HR24L after 2 h and Rad52 was found in HR24L complex after 8 h. Conclusion By showing direct biochemical evidence for the presence of excision and recombination proteins in HR24L complex after radiation damage,the argument that HR24L participates in multiple repair pathway is reinforced.;
