粉防己碱逆转获得性多药耐药小鼠S180肿瘤细胞P_(170)过度表达与调控细胞凋亡相关性研究

摘　　要：　目的:观察粉防己碱对模拟临床化疗PFC方案诱导的小鼠S180细胞多药耐药的逆转作用,明确其对多药耐药逆转的作用及其作用的分子机制,以指导粉防己碱临床逆转肿瘤患者多药耐药,提高化疗疗效。方法:模拟临床PFC方案,分别给予小鼠顺铂3 mg·kg-1,ip,每周 1 次;环磷酰胺和 5 FU各 3 mg·kg-1,ig,每日 1 次,连续 4周,流式细胞术荧光检测观察其对肿瘤细胞膜糖黏蛋白P170、细胞凋亡、细胞凋亡因子Fas,Trail、细胞黏附因子CD54的影响。结果:粉防己碱降低肿瘤细胞耐药基因表达产物P170的过度表达,增加对细胞凋亡因子Fas,Trail表达率,增加耐药S180细胞的凋亡,降低细胞黏附因子CD54的表达。结论:说明粉防己碱逆转肿瘤多药耐药与其对肿瘤多种相关生物分子因子的调节有关。Objective: To observe the effect of tetrandrine on reversion of mice S180's obtained multi-drug resistance tumor cell induced by chemotherapyby PFC. And then discuss the molecular mechanism of it for the use of TCM in clinic to restrain the drug-resistant of chemotherapy,thereby improve the curative effect. Method: By the methods of less dosage of chemotherapy PFC, give the mouse cisplatim 3 mg·kg^(-1) ip, once a week; CTX and 5-FU 3 mg·kg^(-1)ig four weeks,set up the mice models of multi-drug resistance of S180 tumor cell,and then observe the P (170),Fas,CD (54) and apoposis by flow cytometry. Result: Tetrandrine can obviously lower the express of P (170) increase the express of Fas and the apoposis of drug resistant tumor cell. And at the same time it can obviously reduce the express of intercellular adhesion molecule( CD (54)). Conclusion: Terandrine,with the its adjustment of correlated biotic active matter, can intervene the occurrenceof the multi-drug resistance of tumor cells induced by chemotherapy.
