摘  要:目的 评价特异性环氧合酶 2抑制剂对胃癌细胞BGC 82 3的抑制作用及其机理。方法 二种不同浓度特异性环氧合酶抑制剂罗非昔布 (0 .1μmol/L、1.0 μmol/L和 10 μmol/L)和塞来昔布 (5 0 μmol/L、10 0 μmol/L和 15 0μmol/L)干预胃癌细胞株 ,相差显微镜和HE染色观察其形态学变化 ;MTT法检测二者对胃癌细胞的抑制作用 ;ELISA法检测二者干预后胃癌细胞PGE2、VEGF分泌的变化。结果 形态学和MTT法显示 :不同浓度罗非昔布和塞来昔布以时间和剂量依赖性抑制胃癌细胞BGC 82 3增殖 ,同时胃癌细胞分泌的PGE2和VEGF也随着胃癌细胞时间、剂量变化被抑制而分泌减少。结论 特异性环氧合酶抑制剂可通过抑制COX 2代谢通路而抑制胃癌细胞的增殖 ,同时亦可通过抑制VEGF的分泌干预胃癌转移。Objective To evaluate the effects of two different specific cyclooxygenase inhibitors (SCI),rofecoxib and celecoxib,on gastric cancer cell line BGC-823,and to study whether SCI can inhibit cell proliferation and VEGF production interupted by COX-2 metabolic pathway in gastric cancer cell line.Methods According to the plasma peak concentration(PPC),0.1PPC~10PPC rofecoxib and celecoxib were incubated with gastric cancer cell line BGC-823.The suitable concentrations were screened by MTT assay.Changes of cell morphology were observed by invert microscope and HE staoning.The dose-effects and time-effects of the suitable concentrations of rofecoxib and celecoxib on gastric cancer cell line BGC-823 were detected by MTT assay.Changes of PGE_2 and VEGF after intervention with rofecoxib and celecoxib were detected by enzyme-linked immunoadsordent assay(ELISA).Results The suitable concentrations for rofecoxib were from 0.01 μmol/L to 10 μmol/L,and the suitable concentrations for celecoxib were from 10 μmol/L to 150 μmol/L.Under invert microscope,gastric cancer cell line was found fusiform in normal cell,and circular after intervention with rofecoxib and celecoxib.With the concentration added,diaphaneity and cell body became smaller.The same changes were found in HE staining.PGE_2 and VEGF in the supernatant were inhibited with the concentration as proved by ELISA. These manifestations clarified that SCI could inhibit PGE_2 and VEGF with a well dose-effects and time-effects manner in suitable concentrations.Conclusion SCI can inhibit gastric cancer cell line BGC-823 through cyclooxygenase-2-depend pathway,and these effects have a good dose-effects and time-effects manner in a suitable concentrations.At the same time,SCI also can suppress PGE_2 and VEGF in supernatant with a good dose-effects and time- effects manner.The results imply that SCI can suppress angiogenesis and tumor metastasis.
