摘 要:目的 研究Ⅲ型胶原和纤维连接蛋白基因在偏侧咀嚼大鼠磨牙牙周膜表达中的变化 ,探讨偏侧咀嚼对牙周膜改建的分子生物学机理。方法 用 8周龄雄性SD大鼠建立偏侧咀嚼动物模型 ,采用原位杂交的方法检测在偏侧咀嚼 12h及 3、7、14、30d后牙周膜细胞中Ⅲ型胶原和纤维连接蛋白mRNAs表达的动态变化。结果 偏侧咀嚼后的 12h到第 7dⅢ型胶原和纤维连接蛋白mRNAs在牙周膜细胞中的表达主要集中于牙侧和骨侧 ,7d时最明显 ,但在牙周膜中表达的整体强度与正常组没有显著差别 ,14d后二者表达分布与正常组相似。结论 偏侧咀嚼时Ⅲ型胶原和纤维连接蛋白mRNAs表达的分布发生变化 ,这种变化可能与偏侧咀嚼时力增加有关 ,表明纤维连接蛋白和Ⅲ型胶原在偏侧咀嚼时牙周膜组织改建过程中起重要作用。Objective To study the expression of fibronectin and type Ⅲ collagen mRNAs in rat periodontal ligament during experimental unilateral chewing.Methods A model of rat experimental unilateral chewing was established,in which the upper and lower molars on the right side was extracted to remove occlusal force.The rats were perfusion-fixed on the half,3rd,7th,14th and 30th day following the biting-raising.The prepared sagittal sections were processed to study the expression of fibronectin and type Ⅲ collagen mRNAs in rat periodontal ligament by in situ hybridization.Results Compared to the normal rats,following the unilateral chewing the expression level of fibronectin and type Ⅲ collagen mRNAs in the periodontal ligament of the lower first molar had no significant change.But the periodontal cells which expressed both mRNAs were mostly located on the both sides of periodontal ligament.Conclusion The result of expression of fibronectin and type Ⅲ collagen mRNAs in the periodontal ligament following the unilateral chewing suggests that fibronectin and type Ⅲ collagen may play important roles in the process of periodontium remodeling initiated by unilateral chewing.
