TGF-β_1及TGF-βRⅠ信号转导异常与卵巢恶性肿瘤生物学行为的关系

摘　　要：目的　探讨转化生长因子 -β1(TGF -β1)、转化生长因子 -βⅠ型受体 (TGF -βRⅠ )在卵巢癌组织中的表达与其信号转导的临床意义。方法　采用链霉素抗生物素蛋白 -过氧化物酶连接 (S -P)免疫组织化学法检测 85例卵巢良恶性肿瘤组织中TGF-β1、TGF -βRⅠ的表达。 结果　TGF -β1在卵巢良、恶性肿瘤组织中的表达明显高于正常卵巢组织 ,组间有明显差异 (P <0 0 5 ) ;TGF -βRI在恶性组中度阳性至强阳性的表达明显少于良性及正常对照组 ,三组间有明显差异 (P <0 0 5 ) ;TGF -β1、TGF -βRI的异常表达与临床分期有明显关系 ,与年龄和腹水无关 ,TGF -β1与上皮性癌有关 ,TGF -βRI的低表达与癌瘤转移、病理分级呈负相关 (r= -0 2 83 )。结论　TGF -β1、TGF -βRI的表达和信号转导异常与卵巢恶性肿瘤的无止境生长行为有关 ,是评估患者预后的有效参数 ,可能是卵巢恶性肿瘤的重要发病机制之一。Objective To investigate the expression and significance of transforming growth factor-beta1(TGF-β 1),transforming growth factor-beta receptor typeⅠ(TGF-β RⅠ) in human ovarian carcinoma. Methods Inmmunohistochemical SP method was used to detect the expression of TGF-β 1 and TGF-β RI in 85 cases of benign and malignant ovarian tumors. Results The positive rate of TGF-β 1 in benign and malignant ovarian tumors was significantly higher than that in the normal ovarian tissues(P<0.05); The frequency of TGF-β RⅠ middle and strong positive expression was remarkably lower in the malignant tumours than that in the benign ovarian tumors and normal ovarian tissues(P<0.05). The abnormal expression of TGF-β 1 and TGF-β RⅠ was obviously associated with the clinical stages of ovarian carcinoma, but not with patients ages and ascites. TGF-β 1 was related to the histological type of malignant ovarian tumors, and negatively related with pathological staging and metastasis of ovarian carcinoma. Conclusion The abnormal expression of TGF-β 1 and TGF-β RⅠ was associated with the malignant biological behaviors in ovarian tumours, and may serve as a marker for predicting the prognosis of ovarian cancers. TGF-β 1 and TGF-β RⅠ may play an important role in the pathogenesis of ovarian carcinoma.
