摘  要:目的:探讨酸性神经肽1(bovineacidicneuropeptide1,BANP- 1)对吗啡成瘾小鼠脑内γ 氨基丁酸(γ ami nobutyricacid,GABA)及环磷酸腺苷(cAMP)和环磷酸鸟苷(cGMP)含量的影响。方法:48只昆明小鼠随机分为4 组,前10d,吗啡成瘾组、戒断组和BANP -1治疗组每d按100mg/kg注射盐酸吗啡,对照组每d注射生理盐水。第 11d,对照组在注射生理盐水后8h断头处死;吗啡成瘾组在注射盐酸吗啡8h后处死;戒断组在注射盐酸吗啡7.5 h后按4mg/kg注射盐酸纳络酮,0.5h后断头处死;治疗组在注射盐酸吗啡6h后按15mg/kg注射BANP- 1,1.5h 后按4mg/kg注射盐酸纳络酮,0.5h后断头处死。取脑组织匀浆,应用滤纸电泳法测定GABA,应用放射免疫法测 定cAMP和cGMP。结果:吗啡成瘾组各项指标与对照组比较,差异无统计学意义(P>0.05);戒断组cAMP、cGMP 较对照组升高(P<0.05);BANP 1治疗组GABA及cGMP均高于对照组及戒断组(P<0.01)。结论:BANP- 1可能 通过调节吗啡成瘾小鼠脑内GABA及cGMP水平而发挥治疗作用。Aim: To probe the changes of γ-aminobutyric acid (GABA) and cyclic nucleotides (cAMP, cGMP) in morphine addicted mice brain caused by bovine acidic neuropeptide 1(BANP-1). Methods:A total of 48 Kunming mice were allocated into four groups,12 in each group.Three of the groups were injected with hydrochloric morphine at 100 mg/kg for 10 days.The 4th group was injected normal saline as control.On the 11th day,the morphine addicted group,abstinence group, BANP-1 group,and control group were given hydrochloric morphine,hydrochloric morphine and naloxone,hydrochloric morphine and BANP-1,and normal saline ,respectively.Then the mice were killed and the brain tissue was collected and homogenated to extract GABA and cyclic nucleotides for examination.GABA was analyzed by filter paper-electrophoresis method. cAMP and cGMP were analyzed by radioimmunity method.Results:Compared with that of control group,the morphine addicted groups had no significant difference in GABA,cAMP and cGMP(P>0.05). cAMP and cGMP of abstinence group increased significantly (P<0.05).Compared with those of control and abstinence group, GABA and cGMP of BANP-1 group were higher(P<0.01). Conclusion: BANP-1 can exert actions through adjusting the concentrations of GABA and cGMP in morphine addicted mice.
