摘 要:目的研究血管抑素体内抑制视网膜新生血管形成疗效及对血管内皮细胞生长因子(VEGF)表达的影响。方法高浓度氧诱导C57BL/6J小鼠建立视网膜缺血性病变动物模型。将30只小鼠分为5组:正常组;高氧对照组;实验I组(高氧+血管抑素,浓度150μg/μL);实验II组(高氧+血管抑素,浓度200μg/μL);实验III组(高氧+血管抑素,浓度300μg/μL),前两组玻璃体内注射生理盐水2μL,后三组玻璃体内分别注射不同浓度的血管抑素2μL。视网膜切片HE染色,计数各组小鼠视网膜新生血管内皮细胞核数,并加以比较。免疫组织化学法检测各组小鼠视网膜切片VEGF的表达。于鼠龄17d取各组小鼠视网膜及前增生的血管膜,RT-PCR检测VEGFmRNA的表达。结果吸入高氧鼠不论是对照组还是血管抑素治疗组每只眼均可见突出内界膜的新生血管内皮细胞,发生率为100%。高氧+血管抑素(浓度150μg/μL)组、高氧+血管抑素(浓度200μg/μL)组、高氧+血管抑素(浓度300μg/μL)组与高氧对照组相比新生血管细胞核数分别减少了41.96%(P<0.01)、57.40%(P<0.01)、81.73%(P<0.01),在血管抑素治疗的三组中,随着剂量的加大,抑制新生血管形成作用逐渐加强。与正常组相比,高氧对照组、高氧+血管抑素(浓度150μg/μL)组、高氧+血管抑素组(浓度200μg/μl)。To test the efficacy of native angiostatin in suppressing experimental retinal neovascularization induced by oxygen. Mouse models of hyperoxia-induced ischemic retinopathy were established. Angiostatin or NS were injected into the vitreous in 5 groups: normal ,control and various doses. The nuclei of new vessel buds extending from the retina into the vitreous in different groups were counted and compared under the light microscope. Fibrovascular tissues were obtained from 17 day-old mice in different groups. The experssion of VEGF protein was studied by immuno histochemistry. RT-PCR analysis was performed to examine the expression of VEGFmRNA. There were plenty of new vessel buds in the eyes of all mice in hyperoxic condition. The number of the nuclei of new vesselbuds in the murine eyes with injection of angiostatin. They were reduced by 41.96% (P <0.01), 57.40% (P <0.01), 81.73% (P <0.01) respectively. The experssion of VEGF protein and VEGFmRNA were not significantly different in angiostatin groups and control groups. [Conclusions] The proliferation of retinal vessel may be suppressed by using angiostatin. The mechanism of this inhibitory effect probably is not to suppress the expression of VEGF.
