摘 要:目的:建立一种快捷准确的测定薯蓣植物中皂素含量的方法,用以指导皂素生产原料收购及皂素生产过程中质量监控。方法:配制一系列不同浓度的皂素标准品溶液,高氯酸显色后在410nm处测定各溶液紫外吸收度,以浓度对吸收度求出曲线方程。结果:皂素的紫外吸收与浓度具有良好的线性关系,曲线方程:Y=0.0216X-0.0008,在4-45μg/ml浓度范围内曲线相关系数为1.000。我们用该方法对叉蕊薯蓣(DioscoreacollettiiHook.F.),小花薯蓣(DioscoreaparvifloraTing.)和盾叶薯蓣(DioscoreazingiberensisC.H.Wright.)三种植物进行了皂素含量测定,含量分别为2.00%、2.17%、2.29%。测定结果与传统的质量分析法无显著差异。结论:采用紫外吸收光谱法进行皂素含量测定简单易行,线性及重现性良好,可以取代传统的质量分析法作为皂素生产过程中含量测定的新方法。Objective: A fast and accurate method for quantification detemination of Diosgenin in Dioscorea by UV was established. This method can be used to guide purchase the raw material and supervise quanlity of the production line. Methods: Determining a series of Diosgenin alcohol solutions with different density at 410 nm after colorated by perchloric acid, then work out the equation between density and absorb degree. Results: The UV absorb degree and density have a good linear behavior. The correlation coefficient is 1.000 and the equation is Y=0.0216X-0.0008. We use that method to Determine contents of dioscorea collettii Hook.F, dioscorea parviflora Ting, and dioscorea zingiberensis C.H.Wright. Diosgenin content in three kinds of dioscorea is 2.00%, 2.17%, 2.29% respectively. Conclusion: The method is accurate, rapid and with good reproducibility. This method would be used in quality controlling of diosgenin production and finding other diosgenin resource.
