摘 要:目的:通过检测DNA聚合酶β(DNApolymerasebeta,DNApolβ)基因在人肺癌,人正常肺组织中蛋 白水平的表达情况来探讨其在肺癌发生发展过程中的作用和意义。方法:应用免疫组化的方法对67例肺癌, 27例正常肺组织中DNApolβ进行定位及蛋白表达低下率的比较,建立数据库,采用SPSS10.0软件对数据进 行统计学分析,以揭示DNApolβ蛋白表达与肺癌发生的关系。结果:在肺癌组织中,有50.7%(34/67)的人 DNApolβ表达低下;正常肺组织中,有14.80%(4/27)的人表达低下,二者表达低下率存在显著差异,χ2= 10.31,P<0.05,OR=5.92,95%C.I为(2.00,17.51),提示DNApolβ表达低下是肺癌发生的一个危险因素。 且DNApolβ表达与暴露因素如:肿瘤家族史、性别、吸烟状况有关,吸烟可以抑制其表达。肺鳞癌,细胞癌的 低下率远高于其他组织类型,有依鳞癌、小细胞癌、腺癌和其他类型依次降低的趋势χ2trend=9.36,P=0.02。 DNApolβ蛋白表达与TNM分期、细胞分化程度的高低、有无淋巴结转移、3年生存率没有显著相关。结论: DNApolβ表达在肺癌的发生中起着重要的作用,该基因的表达低下可能是肺癌发生的一个分子标志。Objective:To investigate the role of gene DNA pol beta in lung cancer, the level of DNA pol beta protein expression in 67cases of lung cancer and 27cases of nomal lung tissue were tested by immunohistochemistry.Methods: Immunohistochemistry staining was used to test the expression of DNA pol beta protein in lung tumor tissues and normal lung tissues. Results:The DNA pol beta protein expression was low or absent in 34 of 67 (50.7%)lung cancer patients, whereas there were only 4 of 27 (14.8%) normal lung tissues with reduced levels of DNA pol beta protein expression, [χ 2=10.31,P<0.05,OR=5.92, 95%C.I(2.00,17.51)], indicating that the low expression of DNA pol beta was a dangerous factor for lung carcinogenesis. We found that smoking,sex and family history of tumor may inhibit the expression of DNA pol beta gene protein expression;and the status of age, clinicopathologic features such as TNM staging, differentiation, lymph node invading were not correlated with the expression of DNA pol beta, the low expression rate of DNA polyβ-in squamous carcinoma and small cell carcinoma was not only much higher than that in adenocarcinoma and other kinds of histology, moreover, there was a trend of the lowering expression rate by order as in: squamous carcinoma, small cell carcinoma, adenocarcinoma and other kinds of histology.χ 2 trend=9.36,P=0.02. Conclusion: DNA pol beta plays a key role in carcinogenesis of lung,and the low expression of DNA pol beta gene may be used as a valuablely susceptible biomarker of human lung cancers.
