抑癌基因PTEN和P27在非小细胞肺癌中表达与肺癌分化程度及功能预后的相关趋势

摘　　要：目的:探讨抑癌基因PTEN,P27在非小细胞肺癌中表达的意义与预后的关系及其相关性。方法:56例非小细胞肺癌标本及10例正常肺组织均取自泰山医学院附属医院1997-01/1999-03手术切除标本的存档蜡块,应用免疫组织化学方法观察正常肺组织和非小细胞肺癌中PTEN和P27的表达情况。结果:PTEN和P27在肺癌组织与正常肺组织的表达差异具有显著性意义(tau_b=0.341,0.311,P<0.01)。PTEN的表达与有无淋巴结转移有关(tau_b=0.274,P<0.05);与肺癌的分化程度有关(tau_b=0.245,P<0.05),与临床分期有关(tau_b=0.359,P<0.01),与预后有关(χ2=5.22,P<0.05)。P27的表达与肺癌的分化程度有关(tau_b=0.246,P<0.05),与预后有关(χ2=6.30,P<0.05)。PTEN与P27之间的表达无明显的关系(χ2=2.851,P>0.05)。结论:从蛋白水平揭示PTEN的表达与非小细胞肺癌的分化程度,有无淋巴结转移,临床分期以及与肺癌预后有关;P27与肺癌的分化程度及预后有关;PTEN蛋白与P27蛋白之间未发现有明显的关系。AIM:To investigate the relationship between the significance of expressions of anti oncogene PTEN and P27 in non small cell lung cancer(NSCLC) and the prognosis of lung cancer,and probe into the interaction of the two genes.METHODS:The filed paraffin sections of 56 cases of NSCLC and 10 cases of normal lung issues excised by operation were obtained from the Affiliated Hospital of Taishan Medical University from January 1997 to March 1999.Immunohistochemical method was used to determine the expressions of PTEN and P27 in the normal lung issues and NSCLC tissues.RESULTS:The expressions of PTEN and P27 were significantly different in NSCLC and normal lung tissues(tau_b=0.341,0.311,P< 0.01).The expression of PTEN was related with lymph node metastasis(tau_b=0.274,P< 0.05),pathological grade(tau_b=0.245,P< 0.05),clinical staging(tau_b=0.359,P< 0.01) and prognosis(χ 2=5.22,P< 0.05).The expression of P27 was related with pathological grading(tau_b=0.246,P< 0.05) and prognosis(χ 2=6.30,P< 0.05).There was no significant difference between P27 and PTEN(χ 2=2.851,P >0.05).CONCLUSION:The expression of PTEN is related to pathological grade,lymph node metastasis,clinical staging and prognosis of lung cancer.The expression of P27 is related to differentiation and prognosis of lung cancer.PTEN protein is not correlated with P27 protein obviously.
