摘  要:目的:探讨大鼠在模拟5000m高原环境下脑水肿脑组织水通道蛋白4(aquaporin4,AQP4)的表达变化及意义。方法:实验选用Wistar大白鼠56只按随机数字法将大鼠分为正常对照组8只、实验组48只,其中实验组按在模拟高原环境中饲养时间分为饲养4,24,48,72h,2周和4周组,每组8只。在各时相点取大鼠大脑半球组织,测定脑组织含水率,作AQP4免疫组织化学染色和组织原位杂交,用图像分析软件检测AQP4抗体和基因阳性表达相对灰度值,了解AQP4在大鼠大脑的表达变化。结果:饲养4,48,72h组大鼠脑组织含水率明显高于正常对照组(P<0.05~0.01),大鼠在模拟高原环境中饲养2周时含水率逐渐下降,饲养4周组脑组织含水率接近正常对照组。AQP4在脑毛细血管壁及胶质纤维呈阳性表达,正常对照组大鼠AQP4平均相对灰度值(averagerelativegray,ARG)(1.268±0.119)明显低于饲养4h组(1.392±0.098)(t=5.258,P<0.05);饲养24,48和72h组AQP4阳性表达增强,ARG值显著增加(t=2.967~5.652,P<0.01);饲养2周组ARG值有所下降(t=5.239,P<0.05)。AQP4mRNA在胶质细胞核和神经细胞核表达阳性,变化规律与AQP4蛋白变化基本一致,即24,48和72h组阳性细胞数率增加(t=3.045~5.526,P<0.01),阳性染色增强,饲养2周组阳性表达率下降(t=4.687,P<0.05)。结论:AQP4与血脑屏?AIM:To investigate the changes of aquaporin 4(AQP4) expression in the brain tissue of rat models of cerebral edema in the stimulating surroundings of 5 000 m highland, and study its significance.METHODS:Fifty- six Wistar rats were divided into normal control group(n=8) and experimental group(n=48) with the method of random number table,and then the experiment group was subdivided into 4,24,48 and 72- hour,2 and 4- week groups according to the feeding time in the stimulating surroundings of highland with 8 rats in each groups.The tissue of cerebral hemisphere at each time point was obtained,and the ratio of water content was determined,AQP4 immunohistochemical staining and tissue hybridization in situ were performed,and the relative gray level of AQP4 antibody and gene positive expression was detected with the imaging analysis software, so as to investigate the changes of AQP4 expression in the brains of rats.RESULTS:The ratio of water content was obviously higher in the 4,48 and 72- hour groups than in the normal control group(P< 0.05 to 0.01),and it was gradually decreased when the rats were fed in the stimulating surroundings of tableland for 2 weeks, and it got close to that in the normal control group when 4 weeks.The positive expression of AQP4 was observed in the blood capillary dissepiment and gelatinous fiber of brain tissue, the average relative gray(ARG) of AQP4 in the normal control group(1.268± 0.119) was obviously lower than that in the 4- hour group(1.392± 0.098)(t=5.258, P< 0.05),and the positive expressions of AQP4 in the 24,48 and 72- hour groups were enhanced, and the ARG was significantly increased(t=2.967 to 5.652,P< 0.01) while the ARG began to decrease in the 2- week group(t=5.239,P< 0.05).The AQP4 mRNA expressions in the nucleus of glial cells and nerve cells were positive, and its changing law was the almost the same as that of AQP protein,that was,the rate of positive cell number was increased(t=3.045 to 5.526,P< 0.01) and the positive staining was enhanced in the 24,48 and 72- hour gro
