摘 要:目的:探讨不同频率电针预处理(2/5Hz,2/15Hz,2/100Hz)对诱导脑缺血耐受程度是否有差别。方法:实验于2002-01/2002-05在第四军医大学西京医院麻醉科实验室完成。SD雄性大鼠48只,随机分为4组(n=12),即戊巴比妥对照组、电针2/5Hz,2/15Hz和2/100Hz组。戊巴比妥对照组腹腔注射戊巴比妥钠40mg/kg·次,连续5d;电针组在戊巴比妥钠麻醉下根据组别分别用2/5Hz,2/15Hz或2/100Hz频率的疏密波刺激百会穴,30min/d,共5d。最后一次处理24h后,所有大鼠在异氟醚麻醉下用颈内动脉线栓法致右侧大脑中动脉栓塞120min。再灌注24h后,行神经功能障碍评分,并取大脑行TTC染色以测量脑梗死容积。结果:2/5Hz,2/15Hz和2/100Hz电针组神经功能障碍评分中位数分别为1(1-2),0.5(0-1),1(1-2),低于戊巴比妥对照组犤3(2-3)犦(P<0.01),2/15Hz电针组评分明显低于2/5Hz(P=0.008)和2/100Hz组(P=0.002),2/5Hz和2/100Hz两组间评分差异无显著性意义(P=0.843)。电针组的脑梗死容积均明显小于戊巴比妥对照组(P<0.001),2/15Hz组的脑梗死容积明显小于2/5Hz(P=0.02)和2/100Hz组(P=0.004),2/5Hz和2/100Hz两组间脑梗死容积差异无显著性意义(P=0.511)。结论:3种频率的电针预处理均能不同程度地诱导大鼠产生脑缺血耐受,但2/15Hz频率电针诱导脑缺血耐受效果最?AIM: To investigate the difference of electroacupuncture with various frequencies(2/5Hz, 2/15Hz, 2/100Hz) on the degree of induced brain ischemic tolerance in rats. METHODS: The experiment was finished in the Laboratory of Department of Anesthesiology, Xijing Hospital of Fourth Military Medical University of Chinese PLA from January to May 2002. Forty- eight male SD rats were randomly divided into 4 groups (n=12): pentobarbital group, electroacupuncture with 2/5Hz, 2/15Hz or 2/100Hz groups. Rats in pentobarbital group were injected with pentobarbital sodium 40 mg/kg once a day for 5 days. The other three groups received electroacupuncture with 2/5 Hz, 2/15 Hz or 2/100 Hz respectively at the Baihui acupoint 30 minutes a day for 5 days under pentobarbital sodium anesthesia. At 24 hours after the last pretreatment, all rats were subjected to the right middle cerebral artery occlusion(MCAO) for 120 minutes after isoflurane anesthesia. The neurological deficit scores (NDS) were evaluated at 24 hours after reperfusion. The cerebral infarction volume was determined with TTC staining.RESULTS: The median of NDS in electroacupuncture groups with 2/5 Hz, 2/15 Hz and 2/100 Hz groups was 1 (1- 2), 0.5 (0- 1), 1 (1- 2), respectively, was lower than that 3 (2- 3) in pentobarbital group(P< 0.01).The NDS in 2/15Hz group was significantly lower than that in 2/5Hz group (P=0.008) and 2/100 Hz group (P=0.002). There was no significant difference between 2/5 Hz group and 2/100 Hz group(P=0.843) . Infarct volume in electroacupuncture groups was significantly smaller than that in pentobarbital group(P< 0.001). Infarct volume in 2/15 Hz group was significantly smaller than that in 2/5 Hz group and 2/100 Hz group(P= 0.004).There was no significant difference in infarct volume between 2/5 Hz group and 2/100 Hz group(P=0.511).CONCLUSION: Three frequencies of electroacupuncture are able to improve brain ischemic tolerance at different levels. However, electroacupuncture with 2/15 Hz induces the best effect.
