牙龈卟啉菌381胞外膜泡中胰酶样蛋白酶的分离纯化

摘　　要：本实验在分离纯化胰酶样蛋白酶(Trypsin like protease,TLP)的过程中,牙龈卟啉菌(Pg)381胞外膜泡(extracellular vesicles,ECV)经初次GF-HPLC、IE-HPLC层析,分别得到了高活性组分(Ⅱ和Ⅱ-4),Ⅱ-4经过第二次GF-HPLC分离,获得了纯度较高的TLP组分(Ⅱ-4-2).效率明显高于其它分离手段,实验表明:Pg ECV在初步提取后,进一步采用GF-HPLE、IE-HPLC分离,对于纯化TLP具有较好的效果.During the purification of TLP, the enzymes were extracted from ECV of P.gingivalis 381 with HPLC. The result demonstrated that by the first GF-HPLC, IE-HPLC and second GF-HPLC separation, the enzymes were successfully sepcrated from ECV extracts, and shew that the fragments, Ⅱ-4-l(Pg 381) contained the main parts of TLP. This study indicated that separation of enzyme such as TLP with HPLC was a better method than some other separations.
