摘  要:目的:探索核转录因子(NF κB)抑制剂吡咯基二硫氨基甲酸酯(PDTC)对病毒感染搏动心肌细胞的作用及信号转导机制。方法:利用培养的SD乳鼠心肌细胞,分为对照组,CVB3 感染组[心肌细胞感染 coxsackieB3(CVB3)病毒],PDTC干预组(加入PDTC干预后心肌细胞再感染 CVB3 病毒)。采用 MTT法检测细胞活性,采用细胞病变法计算细胞病变;采用Westen blot测定NF kBp65蛋白表达变化。结果:CVB3 感染组与对照组比较,心肌细胞活性明显降低,细胞病变加重;NF kBp65蛋白表达明显高于对照组(P<0.01)。PDTC干预组心肌细胞活性明显低于CVB3 感染组, 细胞病变较 CVB3 感染组轻,NF kBp65 蛋白表达明显低于 CVB3 感染组(P<0.01)。结论:CVB3 病毒在感染早期可导致心肌细胞损伤,NF κB抑制剂可保护心肌细胞免受病毒侵害,NF κB信号通路激活可能为病毒性心肌炎发病机制之一。Objective:Coxsackie B 3 viruses(CVB 3) are considered to be the most common etiologic agents of viral myocarditis. The idea that direct CVB-mediated injury are important in the development of myocarditis . Pyrrolidine dithiocarbamate(PDTC) is the specified inhibitor of nuclear factor kappa-B(NF-κB). Our experiment was to study the anti-CVB 3 ability and Singal transduction of PDTC to cultured beating myocardial cells of CVB 3 infected newborn rat. Method: Neonatal SD mouse cardiac myocyte cultures are prepared with enzymatic digestion, and incubated. CVB 3 group were inoculated with CVB 3; PDTC group were inoculated with PDTC for 30 min, then infected with CVB 3; Control group were treated with same dose DMSO. Western Blot were used to study the expression of p65 protein of NF-κB.Cell viability were analyzed with MTT. Cell cytopathic effect (CPE) was used to compute cell cytopathy. Result:The the expression of p65 protein of NF-κB were higher in CVB 3 group than control groups(P< 0.01). And the protein of NF-κB in PDTC group is lower than CVB 3 group(P< 0.01). In MTT, cell viability in PDTC group are higher than CVB 3 group(P< 0.01). In the CVB 3 group CPE was more obvious than others; And the PDTC groups were observed CPE later and lighter than CVB 3 group; Control group had no cell cytopathy, and the cell viability had no effected. Conclution:As virus attack myocyte, the expression of p65 protein of NF-κB in CVB 3 group was higher than others, which mean CVB 3 infection triggers activation of NF-κB signaling cascade; PDTC can block the viral infection, PDTC has antiviral and protective effect on myocardial cell infected by CVB 3 virus.
