摘 要:目的 了解在河南省流行的人类免疫缺陷病毒(HIV)毒株的流行情况及亚型特征。方法 用套式聚合酶反应(nested -PCR)对28份河南省内的HIV-1感染者外周血单个核细胞(PBMc)中的膜蛋白基因进行扩增,并对C2-V3及其邻区350 -450个核苷序列进行测定和分析。结果 28份样品间的基因离散率为5.72%,与流行于云南的(泰国B)毒株基因离散率 为5.80%,与其它国际亚型,欧美B、A、C、D、F、G、H、J、O亚型基因离散率均在18.05%以上。系统树分析显示,28份毒株与 泰国B亚型聚在一起而远离其它国际亚型。其毒株V3环四肽序列泰国B亚型基因型GPGQ8例占28.57%,欧美B亚型基 因型GPGR13例占46.43%.结论 河南省流行的HIV株属B′亚型,其毒株来自与泰国接壤的云南,在河南流行的时间约在 6~10年,其V3环顶端四肽序列特征以GPGR为主。Objective To investigate the prevalent status and characteristic of subtypes of HIV-1 in Henan province . Methods HIV-1 env gene was amplified by PCR from uncultured peripheral blood mononuclear cells (PBMCs) obtained from 28 HIV-1 seropositive infectors of Henan in 2002.The C2-V3 region (about 450bp)of them was sequenced . Results The innergroup gene divergence of 28 sample is 5.72%,and 5.80% with reference strains of yunnan origin (Thailand B). The sample gene divergence with other subtypes,France B,A,C,D,F,G,H,J,O were over 18.05%. Phylogenetic tree analysis showed 28 Henan B strains clustered closely to Thailand B and far from other international subtypes. The analysis of V3 loop amino sequence of 28 Henan B strains reveals that V3-tip motif of 8 samples (28.57%)is GPGQ of Thailand B subtype gene ,13 samples (46.43%)GPGR of Europe and America B subtype gene .Conclusion There finding suggest the epidemic of subtype B of HIV-1 in Henan province and it is closely related to those identified in yunnan province . It had existed for about 6-10 year .V3 tip motifs of the HIV-1 isolates were mainly GPGR .
