摘  要:本文对苯骈二氮卓类药物的热裂解机制以及其簿层展开后重氮偶合显色的色谱方法测定该类药物及其它含氮药物进行了详细研究。以氯氮 为例,利用TLC和GC-MS法,证实了其主要裂解产物之一为2-氨基-5-氯苯基苯甲酮,以及色谱显色机制为热裂解产物的重氮偶合显色。建立了用于苯骈二氮类及其它含氮药物分析的裂解薄层色谱法。展开系统S_1:氯仿-乙酸乙酯-正丁醇-浓氨水(7∶3∶0.9∶0.05)S_2:四氯化碳-乙醚-正丁醇-浓氨水(7∶3∶l.5∶0.03)。临床毒物分析实样测定结果满意。本法简便、快速、灵敏、专属,最低检测限达0.1μg,适合其它含芳伯胺类药物分析。Study on the pyrolytic mechanism of benzodiazepines and the determination of benzodiazepines and other nitrogen containing drugs by pyrolytic-TLC is discussed in detail in the article. Chlorodiazepoxide was used as an example, it has been proved that one of the main pyrolytic products is 2-amino-5-chlorobenzophenone by TLC and GC-MS. and the colouring mechanism is that the pyrolytic products on the plate are diazotized with NaNO_2plus concentrate HCl and then coupled with naphthylethylenediamine.At the same time.a pyrolytic-TLC method has been established and applied to the analysis of benzodiazepines and other nitrogen containing drugs, the developing systems are:S_1:chloroform-ethyl acetate-n-butanol-concentrate ammonia (7∶3∶0.9∶0.05),S_2:carbon tetrachloride-ether-n-butanol-concentrate ammonia (7∶3∶1. 5∶0.03). Results from the clinical toxic analysis show that the method is simple, rapid,sensitive and specific,it also has better application. suitable for the analysis of other aromatic primary amine drugs.
