摘 要:探讨了外源抗氧化剂超氧化物歧化酶模型化合物MSOD(Cu2C21N14H27Cl13)对人肝癌细胞增殖的影响.采用离体培养的人肝癌细胞SSMC-7721细胞株,接种入96孔板,24h后加入MSOD模型化合物,采用MTT(methylthiazolyltetrazolium)实验,计算肝癌细胞生长抑制率;结果表明,不同浓度组的SOD模型化合物对肝癌细胞的增殖均有抑制作用,而且这种抑制作用呈时间和剂量效应.To investigate the effect of models of superoxide dismutase (MSOD) on the hepatoma cell line. The cultured human hepatoma cell line SSMC-7721 was inoculated into 96-well plates and various concentrations of models of SOD (MSOD) were added into different wells after 24 h. MTT assay was used to calculate the growth inhibition rate of hepatoma cells. Simultaneously the cells were seeded into 24-well plates and taken count of after being dyed. Then the data was analyzed statistically to protract the growth curve. DNA agarose gel electrophoresis was applied to detect the effect of the SOD mimics on the DNA of SSMC-7721 cell lines. The MSOD compound has inhibitory effect on hepatoma cells. And the result shows itself as a domino effect at its duration and various concentrations. The DNA samples from the hepatoma cells treated with the drug presented four well-regulated ladders in the DNA agarose gel electrophoresis.
