摘 要:目的 检测黑色素瘤瘤小鼠高强度聚焦超声(HIFU)治疗前后局部(靶区及边缘)血管内皮生长因子(VEGF)、碱性成纤维细胞生长因子(bFGF)、转化生长因子(TGF) β1和TGF α等血管生长因子的表达,探讨HIFU治疗对局部肿瘤血管生长因子的影响。方法 采用B16黑色素瘤动物模型,实验分3组,即HIFU组、手术组和假照组。于治疗后第3天取各组靶区肿瘤组织(手术组除外)及边缘皮肤组织。采用免疫组织化学法检测VEGF、bFGF、TGF β1、TGF α的表达。结果 假照组靶区肿瘤组织VEGF、bFGF、TGF β1、TGF α表达阳性率分别为80 .0 %、5 0 .0 %、65 .0 %、80 .0 % ,而HIFU组为阴性表达;假照组边缘皮肤组织为阴性表达,而手术组及HIFU组边缘皮肤组织的上述4种血管生长因子的阳性表达率均为10 0 % ,两组差异无统计学意义(P >0 .0 5 )。结论 HIFU治疗能够破坏VEGF、bFGF、TGF β1、TGF α等血管生长因子在肿瘤组织中的表达。Objective To determine the effects of high intensity focused ultrasound (HIFU) on the expression of VEGF,bFGF,TGF-β1 and TGF-α in the local area (center and edge) of B16 melanoma.Methods B16 melanoma bearing animal models were randomly divided into 3 groups:HIFU group ( n =20),surgery group ( n =20) and sham group ( n =20).HIFU (10 mHz,40 W/cm 2) was performed for 10 s each time and cumulative ultrasonic exposed duration was 250-300 s.Central (tumor tissues except that in surgery group) and marginal tissues (skin tissues) were sampled at the third day postoperatively.The expression of VEGF,bFGF,TGF-β1 and TGF-α was detected by immunohistochemical assay in these specimens.Results The positive expression rate of VEGF,bFGF,TGF-β1 and TGF-α in the tumor tissues in the sham group was 80.0%, 50.0%, 65.0% and 80.0% respectively,while that in HIFU group all were negative.These factors in the skin tissues were negatively expressed in sham group,while positively in HIFU group and surgery group.The positive expression rate of VEGF,bFGF,TGF-β1 and TGF-α in marginal tissues in these two groups was 100% ( P > 0.05), while in the sham group,the expression was negative.Conclusion HIFU therapy can suppress the expression of VEGF,bFGF,TGF-β1 and TGF-α in the tumor tissues,meanwhile,it can also increase their expression at the edge of the targeted area.
