摘　　要：[目的 ]探讨反义hTERT表达质粒是否能够抑制HL - 6 0细胞增殖能力及端粒酶活性。 [方法 ]通过脂质体法将反义hTERT表达质粒导入HL - 6 0细胞中 ;以G4 18进行筛选得到阳性克隆 ;以MTT法和体外集落形成实验检测反义hTERT表达质粒对HL - 6 0细胞增殖的抑制作用 ;以TRAP -PCRELISA法来研究反义hTERT对HL - 6 0细胞的端粒酶活性的抑制作用。 [结果 ]转染反义hTERT表达质粒组与空白对照及空质粒组相比 ,细胞增生速度明显较慢 ,克隆形成能力明显下降 (P <0 .0 1) ;反义hTERT对HL - 6 0细胞的端粒酶活性具有明显的抑制作用。 [结论 ]本研究构建的端粒酶反义hTERT表达质粒能够抑制HL - 6 0细胞的体外增殖能力及其端粒酶活性。objectives] To prove if hTERT inhibit the proliferation and the activity of telomerase of HL-60 cells. [Methods] G418 was added into the medium after the plamid was successfully introduced into HL-60 cells by using lipofectactin-mediated DNA transfection; The effects of inhibition on the proliferation of HL-60 cells were detected by MTT and colony forming;The telomerase activity of HL-60 cells was examined by TRAP-PCR ELISA methods. [Results] The growth rate of HL-60 cells transfected by antisense hTERT gene was significantly slower than those of the controls and its colony forming ability decreased significantly (P<0.01). The telomerase activity of the cells was significantly inhibited than those of controls. [Conclusions] The expression of a partial compliment antisense sequence to the mRNA sequence of the protein subunit of telomerase can inhibit the activity of telomerase, inhibit the growth and the ability of colony forming of HL-60 cells.
