野生型p53基因诱导人肝癌细胞系HuH-7凋亡的研究

摘　　要：目的　通过转染野生型p5 3基因作用于肝癌细胞株HuH -7,来研究其诱导肿瘤细胞的凋亡作用。方法　野生型p5 3基因通过脂质体Lipofectamine介导转染人肝癌细胞系HuH 7,96h后,用流式细胞仪进行细胞凋亡分析。结果　流式细胞仪分析细胞凋亡结果:阴性对照组为2 . 8%、转染pUHD 10 3组为4 .1%、转染pUHD- 10 - 3 p5 3组为39. 6 %。结论　野生型p5 3基因可诱导人肝癌细胞系HuH 7发生凋亡。Objective To investigate the effect of apoptosis of tumor cells induced by the wild type p53(wt-p53) gene,human hepatocarcinoma cell line HuH-7 was transfected with wt-p53 gene in our study.Methods While human hepatocarcinoma cell line HuH-7 was transfected with wt-p53 gene by Lipofectamine,the apoptosis in the cells was evaluated by flow cytometry after the treatment for 96 hours.Results 2.8% in the untreated group,4.1% in transfected with pUHD 10-3 group,39.6% in tranfected with pUHD 10-3-p53 group.Conclusion Apoptosis in human hepatocarcinoma cell line HuH-7 could be induced by the wt-p53 gene.
