摘 要:目的 研究热休克蛋白 70 (heatshockprotein 70 ,HSP 70 )在经瞳孔温热疗法(transpupillarythermotherapy ,TTT)小鼠视网膜中的表达。方法 首先对10只C57BL/ 6J小鼠进行TTT照射,激光功率5 0mW ,光斑直径1.2mm ,曝光4 0s,然后分别于激光后2h ,1d、3d ,1周、2周取材,采用免疫组化方法检测视网膜内核层HSP 70的表达情况,计算其阳性率及强阳性率。结果 HSP 70蛋白存在于正常C57BL/ 6J小鼠除视杆、锥细胞外节的视网膜各层细胞,TTT后小鼠视网膜内核层细胞HSP 70表达有一动态变化,2h时阳性表达细胞(阳性率和强阳性率分别为6 6 .85 %±4 .83% ,4 .91%±3.19% )即较正常视网膜明显增加,1d时达到高峰(78.12 %±8.0 9% ,8.15 %±3.4 5 % ) ,然后逐步下降,2周时(3.74 %±1.73% ,0 .5 5 %±0 .2 7% )已基本接近正常水平(2 .91%±0 73% ,0 )。结论 TTT引起的视网膜温度轻微升高可以诱导小鼠视网膜内核层细胞HSP 70的过度表达,从而提高其耐热能力,发挥内源性保护作用。Objective To study the expression of heat shock protein-70 (HSP-70)in the retina of C_(57)BL/6J mouse following transpupillary thermotherapy (TTT).Methods Ten C_(57)BL/6J mouse were anesthetized,and TTT was performed on the dual eyes using an 810nm diode laser (spot size:1.2 mm;duration:40 seconds;power:50 mW).A series of laser pulses were aimed at the posterior pole of the retina.The immunohistochemistry analysis of HSP-70 in the inner layer of the retina was performed at 2 hours,1day,3 days,1week and 2 weeks after laser irradiation respectively.Results HSP-70 immunoreactivity was detected in all layers of the normal retina of C_(57)BL/6J mice,except the outer segment.In laser-treated eyes HSP-70 expression was markedly increased in the inner nuclear layer at 2 hours after TTT,the positive rate and strong positive rate were 66.85%±4.83%,4.91%±3.19% respectively.This increase in HSP-70 expression peaked at day 1 (78.12%±8.09%,8.15%±3.45%) whereafter a decline near to normal (2.91%±0.73%,0) at 2 weeks (3.74%±1.73%,0.55%±0.27%) was detected.Conclusion HSP-70 hyperexpression in the inner nuclear layer can be induced during TTT due to the low temperature increase,and the thermotolerance of these cells induced by HSP-70 lead some proportion of cells toward survival or toward apoptosis instead of necrosis,and thus lead to a decrease of damage of area near to the target.
