摘 要:[目的]探讨γ干扰素(IFNγ)与5氟脲嘧啶(5Fu)联合应用对胃癌MGC803细胞株的抑制增殖、诱导凋亡及凋亡相关基因表达的影响.[方法]采用MTT法、TUNEL法及免疫细胞化学法等检测IFNγ与5Fu联合应用前后胃癌MGC803细胞株的抑制率、细胞凋亡率及增殖细胞核抗原(PCNA)的阳性表达率的变化.[结果]5Fu在5~100mg/L,IFNγ在50~10000U/mL浓度范围内时对胃癌MGC803细胞株均有抑制作用,并表现出明显的浓度依赖性;25mg/L的5Fu与不同浓度的IFNγ联合应用时,显著提高对胃癌MGC803细胞株的抑制率.不同浓度的5Fu和IFNγ分别作用于胃癌MGC803细胞株后,细胞凋亡率均随药物浓度升高而增加,当药物作用时间达48h时,细胞凋亡率达高峰;而PCNA阳性表达率随着药物浓度的升高和作用时间的延长而降低.25mg/L(ID50)5Fu与不同浓度的IFNγ联合应用时,显著提高胃癌MGC803细胞株的细胞凋亡率,明显增强对肿瘤细胞的抑制增殖作用;应用25mg/L5Fu于胃癌MGC803细胞株48h后,p53,bcl2,Cmyc的阳性表达率均明显下降.[结论]5Fu和IFNγ均对胃癌MGC803细胞株具有抑制增殖和诱导凋亡的作用,两种药物联合应用时,其作用明显增强.OBJECTIVE?To study anti-proliferation, apoptosis and expression of apoptosis associated genes after combined treatment of IFN-γ and 5-Fu in MGC-803 gastric cancer cell line.?METHODS?Using MTT ~assay , TUNEL method and immunohistochemical staining.?RESULTS?5-Fu and IFN-γ inhibited cell proliferation in 5~100?mg/L and 50~10?000?U/mL dosage region and it showed dose dependence in MGC-803 gastric cancer cell line, respectively. Proliferation of MGC-803 gastric cancer was significantly inhibited by combined treatment of 25?mg/L 5-Fu with different dose of IFN-γ. APO rate was increased with increasing of dose after treated MGC-903 gastric cancer cells with 5-Fu and IFN-γ, respectively. The APO rate was the highest at 48?h after drug treatment, and it was gradually decreased along with the time of culture. The expression rate of PCNA was decreased along with increasing of dose and time of culture. Combined treatment of 25?mg/L 5-Fu with different dosage IFN-γ can significantly increase APO rate of MGC-803 gastric cancer cell. Expression rate of p?53, bcl-2, and C-myc was significantly decreased after treating MGC-803 cell with 25?mg/L 5-Fu.?CONCLUSION?Treating MGC-803 gastric cancer cell with 5-Fu and IFN-γ may induce anti-proliferation and apoptotic effect. Combined treatment may significantly increase this effect.
