摘 要:目的: 探讨△Np63基因作为食管鳞癌检测指标的可能性. 方法: 50例新鲜食管鳞癌组织,未经放疗或化疗,癌旁组织15例,用免疫组化和RT PCR方法检测组织和外周血△Np63表达情况. 食管癌患者外周血标本21例,用RT PCR方法检测了△Np63mRNA表达情况,胃腺癌12例和10例健康人外周血作为对照. 结果: 食管鳞癌组织△Np63蛋白和mRNA表达明显高于癌旁组织,分别是96%和60% (P<0. 01). 食管癌21例外周血检测中△Np63 mRNA阳性率38%,而胃腺癌和健康人外周血中未见表达. 结论: 食管鳞癌的发生和△Np63过度表达有关,△Np63mRNA外周血检测可以作为一个食管鳞癌的肿瘤标志物.AIM: To detect the expression of △Np63 gene in esophageal squamous cancer. METHODS: Fifty fresh tissue specimens as well as 21 peripheral blood samples of esophageal squamous cancer without chemotherapy and radiation, 15 para-cancerous tissue specimens, 12 peripheral blood samples of gastric adenocarcinoma and 10 peripheral blood samples of healthy volunteer were used to detect △Np63 gene expression using immunohistochemistry and RT-PCR. RESULTS: △Np63 gene expression in cancer tissues was up-regulated compared with that in para-cancerous tissue at both mRNA and protein levels (96% and 60% respectively, P <0.01). △Np63 mRNA was detected in 38% of the peripheral blood samples in esophageal squamous cancer but not in gastric adenocarcinoma and healthy volunteer. CONCLUSION: △Np63 mRNA of peripheral blood can be used as a tumor marker of esophageal squamous cancer.
