摘 要:目的 检测脑星形细胞瘤组织中端粒酶的活性 ,分析其活性变化在肿瘤不同病理类型之间的关系。方法 选用 76例I~IV级脑星形细胞瘤患者的手术切除标本作为实验标本 ;应用端粒重复扩增法 (TRAP -ELISA)分别定量检测不同病理类型星形细胞瘤组织中端粒酶的活性。另取 1 5例正常脑组织标本作为对照。结果 76例脑星形细胞瘤患者中 ,Ⅰ~Ⅱ级者 38例 ,端粒酶阳性者 3例 (7.9%) ;星形细胞瘤Ⅲ~Ⅳ级者 38例 ,端粒酶阳性者 33例 (86 .6 %)。两组端粒酶阳性率之间有显著 (P <0 .0 1 ) ;两组之间 ,端粒酶活性定量水平之间也有显著性差异 (P <0 .0 5 )。 1 5例正常脑组织中未检测到端粒酶活性。结论 端粒酶的活性在脑星形细胞瘤中随着肿瘤活性程度的增高而增加。说明端粒酶作为肿瘤标志物在星形细胞瘤良恶性鉴别、恶性程度评估方面具有重要意义。Objective To investigate the telomerase activity in astrocytoma and its relationship to different histopathological grades of the tumor.Methods Seventy-six tumor samples taken from patients with astrocytoma were used to detect the telomerase activity by TRAP-ELISA method. Another 15 normal brain tissues were employed as the controls.Result In 76 astrocytomas,3 of 38 (7.6%) low-grade astrocytoma(grades Ⅰ-Ⅱ)were detected to be telomerase positive;33 of 38 (86.6%) high-grade astrocytoma (grades Ⅲ-Ⅳ) were positive in telomerase expresson. The positive rates in these two groups were significantly different(P<0.001),and their levels of telomerase activity were also significantly different(P<0.05). The telomerase activity was not detected in 15 normal brain tissues.Conclusion The telomerase activity in astrocytoma increases with advancing histopathological grade of the tumor,which is of- significant importance in differentiating between benign and malignant tumors,and evaluating malignancy of tumors.
