摘 要:背景与目的:化疗耐药与肿瘤细胞表达耐药基因有关,本研究检测人脑胶质瘤中耐药基因的蛋白表达,为今后临床制定个体化化疗方案提供依据。方法:采用免疫组织化学二步法,检测53例人脑胶质瘤标本及6例正常脑组织中P-糖蛋白(P-gp)、肺癌耐药蛋白(LRP)、谷胱苷肽转移酶(GST-π)和六氧甲基鸟嘌呤 DNA甲基转移酶(MGMT)的蛋白表达。结果:在正常组织中仅有1例P-gp阳性表达,其余三个基因的蛋白表达为阴性,在人脑胶质瘤标本中,P-gp、LRP、GST-π和MGMT的蛋白表达的阳性率分别是32.1%、43.4%、 43.4%和50.9%,LRP和GST-π的蛋白表达率与胶质瘤的病理分级之间有相关性。结论:P-gp、LRP、GST-π和MGMT在人脑胶质瘤中均有较高表达率,LRP和GST-π的表达和胶质瘤的恶性程度有关,联合测定多种耐药基因的表达对制定化疗方案和预后判断有参考价值。BACKGROUND & OBJECTIVE: Drug resistance has been associated with drug resistant gene expression in tumors. The aim of this study was to investigate the expression of the four drug-resistance genes, P-glycoprotein (P-gp), lung resistance protein (LRP), gultathione-Stransferase-π (GST-π) and O6-alkylguanine-DNA alkyltransferase (MGMT) in human glioma tissues, and to assess possible associations with clinicopathologic features. METHODS: Tumor specimens from 53 patients were analyzed by immunohistochemistry and data were statistically analyzed. RESULTS: The gene expressions were negative in normal brain tissues except P-gp expression showed positive in one case. The positive rates of the P-gp, LRP, GST-π and MGMT protein expression in glioma samples were 32.1%, 43.4%, 43.4% and 50.9%, respectively. A significant association was observed between high grading and LRP as well as GST-ir protein expression (P< 0.05) in the glioma samples. CONCLUSION: Our data indicate that there are higher positive rates of P-gp, LRP, GST-ir and MGMT protein expression in human gliomas.The higher rate for LRP and GST-π protein expression in the case of malignant glioma suggested that the drug-resistance genes ex- pression proiiling will be uselul lor designing the individual chemotherapies and estimating the prognosis in glioma patients.
