摘 要:目的:探讨大豆异黄酮对60Coγ射线损伤小鼠的防护效果。方法:体重23~29g的80只雌性昆明小鼠随机均分为5组,即正常对照、阳性对照组(即单纯辐照组)及低、中、高剂量大豆异黄酮实验组。照前正常对照、阳性对照组及实验组每天分别以溶剂0.5%羧甲基纤维素钠(CMC-Na)和不同剂量大豆异黄酮(50、100及400mg/kgbw)连续灌胃14d,灌胃至D7,除正常对照组外,其余各组小鼠均接受5Gy60Coγ全身性照射1次,照射后继续灌胃2d及7d后宰杀,取血和肝、脾组织标本。结果:(1)与阳性对照组相比,不同浓度大豆异黄酮可显著提高肝组织抗氧化酶CAT和GSH-Px活性;(2)实验组辐照后短期内肝组织MDA含量一过性升高,但很快即显著降低至正常水平,降低幅度和速度均优于单纯辐照组;(3)照射后期实验组小鼠白细胞上升幅度均高于单纯辐照组,且不同浓度大豆异黄酮能升高受照小鼠脾淋巴细胞转化的刺激指数。结论:大豆异黄酮具有提高受辐照小鼠抗氧化能力和增强免疫功能,有利于受辐照后机体的恢复,能显著减轻辐射对机体的损伤。Objective: To investigate the radiation-protective effect of soybean isoflavone. Methods: 80 normal female Kunming mice weighed 23-29g were randomized into 5 groups:three intervention groups, positive group, and control group. The control group and positive group were given 0.5% sodium carboxymethyl cellulose(CMC-Na),and three intervention groups were given different doses of soybean isoflavone (50, 100, 400 mg/kg) respectively for 14d. The whole body of positive group and intervention groups were given 5.0 Gy 60Co-γ radiation one time at D7 and then the mice were put to death at the D2 and D7 after radiation. Results: Different doses of soybean isoflavone could significantly induce or increase CAT and GSH-Px activities of liver tissue, and MDA content of liver tissue once increased and then decreased significantly compared with positive group. In addition, the increase of WBC was higher than that of positive group, and different doses of soybean isoflavone could increase spleen IOS (index of sensitivity). Conclusion: Soybean isoflavone could increase the antioxidant ability of mice, enhance the immune function, and reduce the radiation injury significantly.
