摘 要:从造纸厂废水排放口纤维垃圾中,分离出一株产生碱性纤维素酶(CMCase)的芽孢杆菌(Bacilussp.)。该菌在碳氮比1∶3、起始pH5.5的条件下产酶量较高。CMCase在pH3~10.6时均有较高活力,以pH4.5~10时活力稳定。Ca++、Mg++等金属离子、EDTA、碱性蛋白酶和洗衣粉等均不影响其活力,最适作用温度为55℃,在60℃下处理1小时,酶活丧失80%。A Bacillus strain No. 1 that produced alkaline cellulase has been isolated from waste fibre in soil near the open of drainage pipeline in the paper making factory in Hangzhou. Its higher production of the alkaline carboxymethyl cellulase (CMC ase ) required a 1∶3 ratio of carbon source to nitrogen, also an initial pH value of 5.5. The CMCase was very active over the broad pH of 3 to 10.6 and was stable in the pH of 4.5 to 10. The CMC ase activity was not affected by Ca ++ , Mg ++ , EDTA, alkaline proteinase and laundry detergents. The CMC ase had an optimum temperature of 55℃, but it was not stable in the temperature more than 60℃. It lost 80% of its activity when exposed at 60℃ for 1 h.
