摘  要:目的研究阳离子脂质体联合转铁蛋白(transferrin ,Tf)介导报告基因虫荧光素酶基因对人肿瘤细胞的靶向转染。方法比较阳离子脂质体Lipofectin、Lipofectin联合含双铁的人转铁蛋白[HTf(Fe) 2 ]介导质粒pDR2 luc转染人肝癌细胞株HepG2 、SMMC772 1、人肾癌细胞株GRC及非洲绿猴肾细胞COS7的转染效率,转染效率通过以液体闪烁计数仪单光子计数法测定荧光素酶活性估计。结果除COS7外,其余3株细胞以Lipofectin联合HTf(Fe) 2 介导pDR2 luc的转染效率均比单用Lipofectin的转染效率高(P <0 0 1) ,其中HepG2 与SMMC772 1中的转染效率增高8倍多,GRC中的转染效率增高4倍多。结论Lipofectin联合HTf(Fe) 2 介导质粒pDR2 luc转染细胞时对人肿瘤细胸株有一定的靶向作用。Objective To study the targeting effect on the human carcinoma cell lines of the cationic liposome plus a receptor ligand transferrin(Tf) mediated firefly luciferase gene.Methods The plasmid pDR_2luc were transfected to human hepatocarcinoma cell lines HepG_2,SMMC7721, human renal carcinoma cell line GRC and simian kidney cell line COS_7,by cationic liposome Lipofectin and Lipofectin plus human transferrin with two molecules of Fe^3+ (HTf(Fe)_2)respectively. The transfection efficiency was evaluated by the activity of expressed luciferase.Results Comparing the Lipofectin-HTf(Fe)_2 complexes with the Lipfctin-pDR_2luc complexes,it was found that except no significance in COS_7(P>0.05), the luciferase activity was at least 8 times higher both in HepG_2 and SMMC7721 (P<0.01) and at least 4 times higher in GRC than that using Lipofectin alone(P<0.01).Conclusion Lipofectin plus HTf(Fe)_2 mediated pDR_2luc delivery has targeting effect on the human carcinoma cells.
