摘 要:目的:鉴于γ-氨基丁酸A受体在临床医学上的广泛应用,从国内外对其受体研究的现状出发,分析其受体的结构与功能的关系以及与其相互作用的有关药物进行讨论。资料来源:应用计算机分别在Medline和Elesiverscience数据库检索与γ-氨基丁酸A受体相关的文献,检索词“GABAAorGABAAben-zadizene”,并限定文献语种为英文。同时在CNKI数据库中检索与γ-氨基丁酸A受体相关的文献,检索词“GABAA”,并限定语种为中文。检索时间:1994-01/2004-08。资料选择:对资料进行初审,选取和γ-氨基丁酸A受体结构以及与其药理学功能相关的文献,然后查找全文,如果该文献所论述的功能或活性与中枢神经系统无关或与临床治疗没有任何联系,则被排除。资料提炼:共收集到36篇和γ-氨基丁酸A受体结构及功能相关的文献,排除24篇中,16篇为重复的同一研究,8篇为Meta分析研究。资料综合:符合标准的12篇文献对受体的结构及活性与受体药理学功能进行了研究,其中作用于该受体的药物有镇静剂安定类、抑制剂巴比妥类、痉挛剂印防己毒素、神经活性甾体类等4大类。结论:深入了解γ-氨基丁酸A受体各亚单位的具体功能,为筛选针对受体亚单位的相关药物提供方法与途径。OBJECTIVE:Because A type γ aminobutyric acid (GABAA) receptor is widely used in clinical medicine,the receptor related drugs and relationship between struct ures and functions of GABAA receptor have been discussed according to the study at home and abroad. DATA SOURCES:Literatures related to GABAA receptor were searched from Medline and Elesiver science database by a computer,keywords is “GABAA or GABAA benzod iazepine',and the language was restricted in English.And simultaneously,they we re searched from CNKI database with the word GABAA in Chinese.Time of search is January 1994 to August 2004. STUDY SELECTION:First trial for data,literatures related to GABAA receptor str uctures and pharmacological functions was selected,and then looking for full tex t.Functions or activities dissertated in literatures without relation to central nervous system and clinical therapy were excluded. DATA EXTRACTION:Thirty six pieces of literatures that are related to GABAA re ceptor structures and functions were found, among which 24 pieces were excluded, 16 pieces were repeated and 8 pieces were Meta analysis study. DATA SYNTHESIS:There are 12 pieces of literatures measuring up in which recep tor structures,functions and pharmacological functions were studied.Four kinds o f drugs including sedative diazepam,inhibitor barbiturate,spasmogen picxition and neuroactivesteoid act on this receptor. CONCLUSION:To study thoroughly the function of GABAA receptor subunits could p rovide techniques and approaches for screening the receptor subunit related anti epilepsy drugs
