uPA和PAI-1与大肠癌侵袭行为的相关性研究

摘　　要：目的探讨uPA、PAI-1在大肠癌中的蛋白表达及其与大肠癌侵袭行为的关系。方法应用免疫组织化学SP法检测53例大肠癌、19例大肠腺瘤、11例正常大肠黏膜标本中uPA和PAI-1蛋白的表达情况。结果uPA和PAI-1蛋白在大肠癌组的阳性表达显著高于正常大肠黏膜组和腺瘤组,并与大肠癌的组织学分级、浸润深度、淋巴结转移、远处转移、Duke's分期密切相关(P<0.05)。uPA与PAI-1在大肠癌中的蛋白表达呈正相关(P<0.05)。结论uPA、PAI-1的共同表达是大肠癌的特征之一,二者的高表达可能预示着大肠癌组织学分化较差、存在侵袭、转移和较差的预后。Objective To investigate the expression of uPA and PAI-1 in colorectal carcinoma .The correlation between their expression and tumor invasion was evaluated. Methods The expressions of uPA and PAI-1 protein were detected in 53 cases of carcinomas, 19 cases of adenomas, 11 cases of normal colorectal mucosas by a streptavidin-peroxidase (SP) technique. Results The positive expressions of uPA and PAI-1 protein in colorectal carcinomas were significantly higher than those in normal colorectal mucosas and adenomas, and were related with histological grades, the depth of colorectal carcinoma invasion, lymph node metastasis, distant metastasis and Duke's stage (P<(0.05)). A positive correlation was observed between the expression of uPA and that of PAI-1 (P<(0.05)). Conclusion The co-expression of uPA and PAI-1 was one of the characteristics of colorectal carcinoma, and was associated with poorly histological grades, cell migration , tumor metastases and poor prognosis.
