芋螺毒素RNA的提取及其cDNA合成的研究

摘　　要：采用不同的RNA提取方法,从7种海南产芋螺的毒管中分离总RNA,用两种反转录酶分别合成cDNA,再以合成的cDNA为模板进行PCR扩增。经综合比较,建立了简便快速的芋螺毒素RNA提取及其cDNA合成的优化方法。为进一步分离克隆芋螺毒素新基因提供参考。Two methods of different RNA extractions from Conus venom tube and toxin gland of seven cone snail species were performed. ThermoScript~ TM RNase H~ - Transcriptase and AMV Transcriptase were used to synthesize cDNA separately. The cDNAs were used as templates to amplify conotoxin genes by PCR.The optimized RNA isolation and cDNA synthesis methods were obtained according to RT-PCR results, which would be the basis for cloning of new conotoxin genes.
