摘 要:目的研究白血病患者的骨髓血管新生程度和骨髓液血管内皮生长因子(VEGF)水平的相关性。方法采用EnVison免疫组化法,用兔抗人vWF(vonWille蛳brandfactor)多克隆抗体标记骨髓内皮细胞;采用ELISA法检测骨髓液中VEGF的含量。结果骨髓MVD在各实验组均明显高于正常对照组(P<0.05)。骨髓液VEGF水平在AML、CML组明显高于正常对照组(P<0.01),ALL组与正常对照组比较差异无显著性(P>0.05)。对各白血病组的MVD和VEGF含量进行了相关性分析,结果表明在AML或CML组中VEGF水平和MVD呈正相关性(分别为r=0.648,P<0.05;r=0.733,P<0.05)。结论白血病患者骨髓微血管数增多,提示血管新生在白血病发病中有重要作用,VEGF可能在一定程度上对血管新生有促进作用。Objective To investigate the relation between the angiogenesis of the bone marrow and the level of vascular endothelial growth factor(VEGF) in the bone marrow fluid in leukemia patients. Methods Immunohistochemical technique (EnVison) was employed to stain microvascular endothelial cells in each bone marrow specimen section with anti- human von Willbrand factor. ELISA was employed to measure the level of VEGF in the bone marrow fluid. Results MVD of AML group, ALL group and CML group were significantly higher than those in nomal controls(P <0.05). The VEGF level in the bone marrow fluid of AML group and CML group was enhanced significantly compared with that of normal control(P < 0.01), but VEGF were not significantly increased in ALL(P > 0.05). By analysing the correlations between the level of VEGF and MVD in all groups. We found a direct correlation between the levels of VEGF and MVD in AML group(r = 0.648, P < 0.05) or CML group (r =0.733, P < 0.05). Conclusions Increased MVD in the bone marrow of leukemia patients suggest that angiogenesis might play an important role in the pathogenesis of leukemia. VEGF might promote angiogenesis in bone marrow partly.
