摘 要:在肿瘤发生与发展的过程中,由于正常的基因调控紊乱,可导致细胞信号传导网络的异常,肿瘤细胞内较常出现异常的通路包括受体酪氨酸激酶通路、磷脂酰肌醇3激酶通路、Wnt通路和Rb信号通路等。传统肿瘤化学治疗的主要目的是杀伤癌细胞,但此类细胞毒性药物难以避免选择性差和毒副反应大等缺点。随着分子肿瘤学和细胞分子生物学的进步,采用小分子抑制剂、单克隆抗体和反义核苷酸等选择性针对信号传导通路中某些特定靶分子进行干预治疗,抑制细胞过度增生、上调细胞凋亡以及通过构建重组细胞系,根据虫荧光素酶的表达水平开发出一种实用的高通量抗肿瘤药物筛选体系,已经给肿瘤临床治疗带来希望,并已成为有前景的新领域。Regulatory disturbance of genes often causes abnormality of cell signal transduction network in the process of tumor development. There are often abnormal pathways in tumor cells which include receptor tyrosine kinase pathway, phosphatidylinositol 3 kinase pathway, Wnt pathway,Rb pathway etc. The main aim of traditional tumour chemical therapy is to kill and wound cancer cells, but this kind of cytotoxicity drugs are hard to avoid weak selectivity,big side effect etc., shortcomings. Along with the advancement of molecular oncology,cellullar and molecular biology, treameats of aiming at certain target molecules in the cell signal transduction by small molecule inhibitor,monoclonal antibody,antisense oligonucleotides etc., suppressing cell excess proliferation or up-regulating apoptosis and developing a kind of practical flux antitumor drug screening system according to expressional level of luciferine enzyme through reconstituted cell line constructed have brought expectation to clinical therapy and become hopefull new domain.
