摘  要:目的建立高效液相色谱法测定多糖相对分子质量和含量的方法;分析太子神悦胶囊中多糖的相对分子质量分布和含量测定。方法采用SephadexG-100凝胶柱纯化分离不同相对分子质量的多糖组分,用TSK-GELG3000SW,7.8mm×300mm色谱柱,2410示差折光检测仪,测定其相对分子质量分布和含量。结果太子神悦胶囊中相对分子质量17.0×103-27.0×103的多糖(JNTP2)的含量为(0.805±0.01)%;相对分子质量分布为Mr:24×103;Mn:17×103;Mp:27×103;Mr/Mn为1.4结论建立了简便、快捷、精密度高的凝胶色谱方法;较为准确的测定了太子神悦胶囊中多糖的含量和相对分子质量分布,为制定太子神悦胶囊多糖的质量标准提供依据。OBJECTIVE: To develop a simple size-exclusion high-performance liquid chromatographic (SE-HPLC) method for determining the average molecular mass range, polydispersity and the content of polysaccharides in Taizishenyue capsule. METHODS: Polysaccharids were seperated on a TOSOH TSK-GEL G3000SW HPLC column with 0.1 mol&middotL-1 NaNO3 buffer as a mobile phase, Refractive index detector were used. RESULTS: A good linearity was obtained with the correlation coefficients over 0.999 1. The average recoveries of polysaccharides ranged 69.42%-70.43% and relative standard deviations(RSDs) were below 5%. The content of JNTP2 was (0.805 ± 0.01)%. The molecular weight distribution of JNTP2 were Mr: 24 × 103, Mn: 17 × 103, Mp: 27 × 103 and Mr/Mn: 1.4. CONCLUSION: The method is simple, rapid, precise and has been used for the analysis of the polysaccharids from Taizishenyu capsules.
