摘  要:本文采用整体犬急性心肌缺血-再灌注模型以探讨穿心莲提取液减轻钙超负荷的机制。结扎冠脉左前降支上1/3处90min后再灌注60min较持续结扎150min,缺血中心区心肌细胞内Ca2+增加(P<0.05)、Na+明显增加(P<0.01),心肌细胞膜Ca2+-ATPase活性明显降低(P<0.01),心肌组织MDA含量显著增高(P<0.01)。而在再灌注前45min静脉给予穿心莲提取液,则见其缺血中心区心肌细胞内Ca2+降低(P<0.05)、Na+明显降低(P<0.01),心肌细胞膜Na+-K+-ATPase、Ca2+-ATPase活性明显增加(P<0.01)。心肌组织MDA含量显著降低(P<0.01)。说明穿心莲提取液减轻心肌缺血-再灌注过程中钙超负荷与减轻氧自由基危害、保护心肌细胞膜ATPase活性、降低钠超负荷有关。Mechanism of APN in alleviating the Ca ̄2+-overloading in dog model during the process of ischemic reperfusion was studied.In comparison with the sustained ischemic group,the parameters in reperfusion ischemic group showed:In central ischemic region of myocardial cell Ca ̄2+ increased (P<0.05),Na ̄+ increased markedly (P<0.01),Ca ̄2+-ATPase activity dropped markedly (P<0.01),MDA increased significantly (P<0.01),whereas in the APN pretreated group,Ca ̄2+ in relevant area reduced(P<0.05),Na ̄+ decreased significantly(P<0.01),and the activity of Ca ̄2+-ATPase and Na ̄+-ATPase increased markedly(P<0.01),MDA decreased significantly(P<0.01).These data indicate that APN may improve the activity of Sarcolemma ATPase in alleviating the Ca ̄2+ and Na ̄+-overloading by decreasing the harmful effect of O_2 free radicals.
