摘  要:目的 探讨昆明山海棠胶囊的遗传毒性及生殖系统毒性。方法 根据《中药新药研究指南》,进行该制剂的特殊毒理实验研究。结果 昆明山海棠胶囊无移码突变和碱基置换效应;剂量低于2 5 0g·kg-1时,小鼠骨髓细胞姊妹染色单体交换频率(SCE)正常,高于此剂量,则可增加SCE频率,且使嗜多染红细胞微核率增加,并呈剂量依赖性;显性致死试验阳性;雌鼠均不能受孕;精子畸形率显著增高,随剂量增加而精子畸形率增高;对孕鼠重量、活胎重量及平均胎仔数等各项指标均无影响。结论 昆明山海棠胶囊具有低、中度的遗传毒性和一般生殖毒性,而无胚胎毒性。Aim To study the mutagenicity and reproductive toxic i ty of Kunmingshanhaitang(KMSHT) capsule.Methods Special toxicol ogical experiments of the preparation were carried ant according to 《A Research of New Herb》.Results KMSHT capsule had no effects of frame sh ift mutation and base replacement.The frequency of SCE in mouse bone marrow cell was normal when the dose was lower than 250g·kg -1.The frequency of SCE a nd the percent of PEC-micronucleus increased when the dose was higher than 250g ·kg -1,and showed dose-relevant trend.Dominant lethal test was positive.N o female mice involved was able to become pregnant.Sperm malformation rate was i ncreased markedly along with the increase of the dose.There was no effect on the weight of pregnant mice,the weight of vivifetus and the average amount of fetus .Conclusion KMSHT capsule had low to middle grade of genetic to xicity and general reproductive toxicity,but no embryo toxicity.
