摘  要:目的:观察葛根素对兔肺缺血再灌注损伤中细胞凋亡的影响。方法:复制兔单侧肺缺血再灌注损伤模型,随机分为三组,每组10只:对照组(C组)、缺血再灌注组(IR组)和葛根素组(Pur组)。对比观察各组血浆超氧化物歧化酶(SOD)活力、一氧化氮(NO)及丙二醛(MDA)含量、肺湿干重比(W/D)、肺泡损伤数定量评价指标(IQA)及肺组织细胞凋亡指数(AI)。结果:再灌注后各时间点IR组与C组比较发现,SOD活力、NO含量明显降低(P<0.01),MDA、W/D、IQA、AI明显升高(P<0.01),Pur组与IR组相比较发现,SOD活力、NO含量均明显升高(P<0.01),MDA、W/D、IQA、AI不同程度地有所降低(P<0.05)。结论:葛根素通过抗氧化与提高NO含量,可抑制再灌注后肺组织细胞凋亡,从而减轻肺损伤。objective: To investigate the effect of puerarin on pneumocyte apoptosis in vivo lung ischemia-reperfusion injury in a rabbit model. Methods: Rabbits used for unilat- eral lung ischemia-reperfusion model were randomly divided into three groups(10 rabbits in each group): control group (C group), ischemia-reperfusion group (IR group), and puerarin group (Pur group). The activity of plasma superoxide dismutase (SOD), the content of plasma nitric oxide (NO) and malondialdehyde (MDA), the wet to dry weight ratio of lung tissue (W/ D), the index of quantitative assessment of histologic lung injury (IQA) and apoptosis index (AI) of lung tissue cell were measured respectively in different groups. Results: The activity of SOD and the content of NO were significantly lower in IR group than that in C group (P < 0. 01); while the content of MDA, the value of W/D, IQA , AI were evidently higher ( P < 0. 01). Pur group in contrast with IR group showed that the levels of SOD and NO were high ( P < 0. 01) , however, the values of MDA, M/D, IQA, and AI were low in different degree ( P < 0. 05). Conclusion: Puerarin can suppress apoptosis of lung tissue cell in the reperfusion phase, which results in the obvious attenuation of the injury of pneumocyte because of it's role against the reactive oxygen spieces and for NO.
