转移与无转移肝细胞癌患者血清蛋白质指纹图谱的比较

摘　　要：目的比较转移与无转移肝细胞癌(HCC)患者的血清蛋白质指纹图谱,筛选与肝癌转移相关的蛋白质.方法术前收集84例HCC患者(有转移45例,无转移39例)和30例健康成人的血清,采用弱阳离子交换蛋白质芯片为检测介质,经表面加强激光解吸电离-飞行时间-质谱(SELDI-TOF-MS)测定,得到蛋白质指纹图谱并运BioMarker Wizard软件分析比较.结果在质荷比(m/z)1 500～30 000范围内,比较有转移HCC与健康成人的血清,检测出的146个蛋白质峰中,69个蛋白质峰差异有统计学意义(P<0.05),比较无转移HCC与健康成人的血清,60个蛋白质峰差异有统计学意义(P<0.05),差异蛋白质峰中57个为有转移与无转移HCC的共有蛋白峰;单独比较有转移与无转移HCC血清,检测出的146个蛋白质峰中仅有11个蛋白质峰差异有统计学意义(P<0.05).结论有转移与无转移HCC患者血清蛋白质之间有差异,但远较其与健康成人血清之间差异小;血清蛋白质指纹图谱中的差异蛋白质可能与肿瘤的发生和转移有关.Objective To investigate the serum proteome fingerprint in hepatocellular carcinoma (HCC) patients with or without metastasis and to screen serum proteins with relation to metastasis.Methods Serum samples were collected from 45 HCC patients with metastasis,39 HCC patients without metastasis and 30 healthy adults.Special serum protein or peptide fingerprint was determined by surface enhanced laser desorption/ionization-time of flight -mass spectrometry (SELDI-TOF-MS) measurement after treating the sample onto weak cation exchange (WCX2) protein chip for each case.The obtained data were analyzed by BioMarker Wizard software.Results Within the range from 1 500 to 30 000 at the m/z value,146 protein features were stably detected in the serum protein pattern,among which,69 and 60 proteins had statistically significant difference between HCC patients with metastasis and healthy adults,HCC patients without metastasis and healthy adults,respectively,whereas 57 proteins were shared by HCC patients with or without metastasis,only 11 proteins had statistically significant difference between patients with and without metastasis.Conclusion Statistical difference exists in serum proteins between HCC patients with and without metastasis,but the difference is less than that between healthy adults and HCC patients with or without metastasis;These differential proteins may be related with the carcinogenesis and metastasis of HCC.
