摘  要:目的:探讨不同时程急性惊厥大鼠脑电图及大脑皮质及海马中谷氨酸脱氢酶活性的变化与惊厥的关系。方法:实验于2004-10/12在河北医科大学基础医学研究所生物化学研究室完成。选用成年健康雄性SD大鼠26只,随机分为急性惊厥模型组(n=20),腹腔注射60mg/kg戊四氮致痫复制大鼠急性惊厥模型,造模后又分急性惊厥发作后0,4,24h和7d4个时程阶段组,每组5只。生理盐水对照组(n=6):腹腔注射等体积生理盐水。观察大鼠行为学表现及脑电图的变化,应用DGKC速率法检测大鼠皮质和海马中谷氨酸脱氢酶的活性。结果:①急性惊厥组大鼠注射戊四氮后5~15min均出现Ⅳ~Ⅴ级癫痫样发作,可持续达1h,之后在行为学上的表现逐渐安静和趋于正常。②生理盐水对照组大鼠脑电图为正常α节律,急性惊厥组大鼠脑电图呈现高幅棘-慢、尖-慢波以及多棘、多尖综合波。③谷氨酸脱氢酶活性:急性惊厥模型组大鼠大脑皮质及海马0h犤(21510.97±1040.86),(17542.51±1107.39)nkat/g犦,24h犤(12625.52±2066.04),(12091.08±709.74)nkat/g犦;7d犤(14253.18±1099.98),(13826.10±797.60)nkat/g犦,与生理盐水对照组相比,均显著降低犤(25246.99±2211.15)nkat/g,(21635.72±2154.54)nkat/g,P<0.05犦。急性惊厥后7d有所回升,但仍未回到正常水平。结论:戊四氮?AIM:To study the changes of electroenciphalogram and activity of glutamate dehydrogenase in cortex and hippocampus in rats with acute convulsion at different time points, and the relation with convulsion. METHODS:The experiment was finished in the Department of Biochemistry,Institute of Basic Medicine of Hebei Medical University,from October to December 2004.A total of 30 rats were divided randomly into acute convulsion group(n=20) and saline group(n=6).The convulsive models induced by abdominal injection of pentylenetetrazol(PTZ)in rats were divided randomly into 0,4,24 hour and 7 day groups(n=5) after episode .Rats in saline group were injected with the same dose of saline.The behavior and electroenciphalogram were observed.The activity of glutamate dehydrogenase in cortex and hippocampus of rats were detected with DGKC velocity techniques. RESULTS:①After administration of PTZ for 5-15 minutes,rats showed seizure of Ⅳ-Ⅴgrade regularly, which even lasted for 1 hour.②Rats in saline group showed αrhythm waves;but acute convulsion group indicated high amplitude spike slow waves,spike slow waves, sharp waves or spike waves.③The activity of glutamate dehydrogenase in cortex and hippocampus of rats in acute convulsion group[0 hour:(21 510.97±1 040.86),(17 542.51 ±1 107.39) nkat/g;24 hours:(12 625.52±2 066.04),(12 091.08 ±709.74) nkat/g;7 days:(14 253.18±1 099.98),(13 826.10 ±797.60) nkat/g] was significantly decreased as compared with that in saline group[(25 246.99±2 211.15),(21 635.72±2 154.54) nkat/g](P< 0.05).The activity of glutamate dehydrogenase increased at 7 days after acute convulsion,but which still did not reached to the normal level. CONCLUSION:Electroenciphalogram of convulsive rats induced by PTZ indicates characteristic changes of waves.But the changes of activity of glutamate dehydrogenase are not the same as the behavior of rats.The decreased activity of glutamate dehydrogenase affects the metabolism of glutamate,so as to induce the excitatory toxicity in brain tissue,which
