摘 要:目的:确定转化生长因子β1(transforminggrowthfactor-beta1,TGF-β1)基因启动子区的多态性和慢性阻塞性肺疾病(chronicobstructivepul-monarydisease,COPD易感性的关系。)方法:选择2004-02/08在四川大学华西医院住院的稳定期COPD患者84例为研究组,男62例,女22例;同期选择与患者组性别、年龄匹配的无血缘关系的四川成都地区汉族健康人97例为对照组,男71例,女26例。应用PCR-RFLP方法检测了两组研究对象TGF-β1基因启动子区的多态性。结果:-509C>T不同基因型在研究组和对照组之间的分布差异有显著性意义(P<0.05)。-509T等位基因在研究组中的频率(19.6%)也显著低于对照组(31.4%)(P<0.05)。在重度吸烟者中,-509C>T不同基因型及等位基因在研究组和对照组之间的分布差异有显著性意义(P<0.05)。结论:TGF-β1基因-509C>T多态性与COPD的发生相关联,-509T等位基因可能是COPD的保护因子之一。AIM:To determine the correlation between polymorphism of transforming growth factor beta 1(TGF β 1) promotor region and susceptibility of chronic obstructive pulmonary disease(COPD).METHODS:Eighty four stable COPD inpatients(study group, 62 males and 22 females),who were hospitalized in West China Hospital of Sichuan University, and 97 age and gender matched healthy Han subjects(control group,71 males and 26 females) without blood relationship in Chengdu area of Sichuan province were selected from February to August 2004.The polymorphism of target fragments of TGF β 1 gene promoter were amplified and analyzed by using the technique of polymerase chain reaction restriction fragment length polymorphism(PCR RFLP) in both groups.RESULTS:Different genotypes of the polymorphism - 509 C >T distributed significantly different between the study group and control group(P< 0.05). The frequency of the - 509T allele was significantly lower in the study group(19.6% ) than in the control group(31.4% ) (P< 0.05). The distributions of - 509 C >T different genotype and allele were remarkably different between the study group and control group(P< 0.05). CONCLUSION:The TGF β 1 gene - 509 C >T polymorphism may be associated with the occurrence of COPD,and the - 509T allele is possibly one of the protective factors from COPD.
