鬼臼乙叉甙对星形胶质细胞瘤表达β1,4-半乳糖基转移酶的影响

摘　　要：目的　研究鬼臼乙叉甙(Etoposide ,又称VP16)影响人星形胶质细胞瘤SHG 44细胞周期过程中,与N 糖链合成有关的β1,4 半乳糖基转移酶Ⅰ、Ⅱ、Ⅴ(β- 1,4 -galactosyltransferaseⅠ、Ⅱ、Ⅴ,β- 1,4- GalT- Ⅰ、Ⅱ、Ⅴ)表达变化。方法　首先应用流式细胞仪技术分析VP16处理SHG 44细胞过程中细胞增殖情况的变化,再采用NorthernBlot方法分析处理过程中的SHG 44细胞中β- 1,4 -GalT- Ⅰ、Ⅱ、Ⅴ的表达变化。结果　流式细胞仪检测表明,细胞培养基加入12 0 μmol/LVP161～6h后能引起SHG 44细胞的S期阻滞。NorthernBlot显示,VP16作用1～6h过程中,β- 1,4 GalT -Ⅰ、Ⅴ表达无显著变化。而β- 1,4- GalT -Ⅱ表达在此过程中则有升高趋势。结论　VP16能够引起星形胶质细胞瘤细胞SHG 44细胞周期改变过程中,β- 1,4 -GalT- Ⅰ、Ⅴ的表达无变化,而β- 1,4 -GalT- Ⅱ的表达明显升高。提示VP16引起肿瘤细胞周期改变,可能通过影响β- 1,4- GalT- Ⅱ的表达,引起有关蛋白质的糖基修饰而实现的。Objective The purpose is to investigate the expression of β1,4-galactosyltransferase Ⅰ?Ⅱ and Ⅴ(β-1,4-GalT-Ⅰ、Ⅱ and Ⅴ) in human astrocytoma.Methods Cell cycles of astrocytoma SHG-44 treated with Etoposide were determined by fluorescence-activated cell sorting(FACS).The changes of β-1,4-GalT-Ⅰ、Ⅱ and Ⅴ in treated astrocytoma cells were studied by Northern Blot.Results It was discovered that cell cycles of astrocytoma SHG-44 were changed after treated by Etoposide,and during this course,the expression of β-1,4-GalT Ⅱ mRNA was higher while those of β-1,4-GalT Ⅰ and Ⅴ mRNA had no obvious changes.Conclusion The increased expression of β-1,4-GalT-Ⅱ in astrocytoma SHG-44 treated with Etoposide suggested the β-1,4-GalT-Ⅱ may take part in the regulation of cell cycles of SHG-44 treated by Etoposide through the modified glucoprotein.
