摘 要:以标准SDS法、CTAB法、改良SDS法、CTAB法对小麦基因组DNA提取质量进行了比较研究。结果表明,标准方法所提DNA浓度和纯度都较好,但耗时、设备条件要求高;改良方法所提DNA浓度和纯度虽不如标准法,但依然符合实验要求,而且省时、经济、易操作。改良CTAB法提取DNA质量好于改良SDS法,改良方法尤其改良CTAB法是一种适合小麦基因组DNA提取的方法。此方法可用于教学实验和科研中对大量材料DNA的提取,其质量可满足对DNA质量要求较高的实验需要,如AFLP技术。This study aimed at comparing the different methods of DNA extraction from wheat. The results showed that the DNA quality extracted by standard operation was good, but it spent so much time and money. The DNA concentration and purity extracted by improved method were not as good as standard, but it also met the DNA requirements of major molecular research . The improved CTAB extraction method which was designed by our research group is suitable to extract the high quality DNA from wheat. Facile operation can save time and be economical. The quality of DNA extracted by improved CTAB method is so good that it fits for AFLP experiment.
