黏附分子CD44v6和E-cadherin结肠癌小鼠表达的实验研究

摘  要:[目的]探讨正常小鼠组织与结肠癌(C26)小鼠黏附分子CD44v6和E-cadherin表达的关系。[方法]应用SP免疫组织化学方法检测结肠癌小鼠和正常小鼠组织中CD44v6和E-cadherin的表达情况,以组织标本的平均密度(MOD)表示。[结果]结肠癌小鼠组织中CD44v6的MOD为0.1103±0.0020,而对照组为0.0868±0.0187;结肠癌小鼠中CD44v6表达水平比正常小鼠对照组明显增强(P<0.01);结肠癌小鼠组织中E-cadherin的MOD为0.1100±0.0034,正常对照为0.1426±0.0015,结肠癌小鼠比正常小鼠明显降低(P<0.01),且CD44v6表达水平与E-cadherin表达水平呈负相关(P=0.000)。[结论]结肠癌的生长、侵袭可能与黏附分子异常表达相关,检测CD44v6和E-cadherin表达可作为判断结肠癌生长和侵袭倾向的参考指标。To investigate the relationship of CD44v6 and E-cadherin expressions between normal tissue and colonic cancer tissue in mice. SP immunohistochemical method was used to detect CD44v6 and E-caclherin expression in different tissue, respresented by Mean Optical density(MOD). The MOD of the CD44v6 were 0.1103±0.0020 in colonic cancer group and 0.0868±0.0187 in control group, but the MOD of the E-cadherin were 0.1100±0.0034 in colonic cancer group and 0.1426±0.0015 in control group. The CD44v6 expression level in mice colonic cancer tissue was significantly higher than that in mice normal colonic tissue(P<0.01). The E-cadherin expression level in mice colonic cancer tissue was lower significantly than that in mice normal controls(P<0.01). Moreover, there was a negative correlation between the CD44v6 and E-cadherin expressions in colonic cancer group(P=0.000). [Conclusion] The growth and invasion of the colonic cancer may be related to the abnormal expression of adhesive molecule. CD44v6 and E-cadherin may be used as a marker of progression for colonic cancer.
