摘 要:应用激光多普勒电泳技术比较了在42.5℃加温治疗后MA737乳腺癌和相应正常乳腺细胞的膜电荷效应,并观察了利多卡因对细胞膜热损伤的扩大作用。结果表明,肿瘤细胞随42.5℃加温时间,从0’,30’,60’,90’到120’的延长,EPM值下降,分别为2.839,2.460,2.265,2.076和1.870μm/sec/v/cm,而加温对正常细胞的表面电荷没有明显影响。表明肿瘤细胞比正常细胞有较高的热敏性。这两种细胞对温度反应的差异,与各自细胞膜治疗前的物理状态不同有关。局麻药利多卡因可进一步降低加温诱发的肿瘤细胞迁移速度的下降,其与肿瘤生长抑制关系作了初步分析。Heating (42.5℃) effect on cell membrane charge of MA737 mammary carcinoma and normal breast cells were compared using techenique of laser Doppler electrophoresis and the enhancement of thermal damage on tumuor cells by lidocaine were observed. The results showed that the values of EPM were decreased being 2.839, 2.460, 2.265, 2.076 and 1.870um/sec/v/cm for 0', 30', 60', 90' and 120' after heatihg. However, there was no significant effect observed on surface charge of normal cells after heating, the results suggested that the thermal sensitivity on tumuor cells was higher than on normal cells, as a result of difference of their respective physical phase before treatment. Besides, a reduced mobility of MA737 cells could be observed after heating and lidocaine addition.The correlation between the decrease of MA737 cell mobility and the slowing of tumor growth was also studied.
