摘 要:基于补体介导的溶血原理,建立了测定血型物质含量的溶血抑制试验。分型血清抗体与血球结合后,在补体的作用下可以溶解血球而释放血红蛋白,通过测定上清中血红蛋白含量求出所测样品中的血型物质含量。该方法的灵敏度(0.078μg/ml)比常规的血凝抑制试验(0.3125~0.165μg/ml)高4~8倍,重复性好,简便易行,可用于测定生物制品中血型物质的含量。A new method (hemolysis inhibiton test) for the determination of blood group substance content was established based on the complement mediated hemolysis principle. Under the influence of complement, typing anti-serum combined with blood cells could dissolce blood cells and release homoglobin. Blood group substance content Could be calculated out by determining homoglobin content in supernate. The method was simple and easy to handle, and showed good repetition. The sensitivity of it was 4~8 folds higher than that of routine inhibition test. It proved that the method was suitable for the determination of blood group substance content in biological products.
